目的:建立维吾尔医异常黑胆质型肝癌病证大鼠模型,探讨信号转导及转录活化因子3(STAT3)基因在异常黑胆质型肝癌病证发生发展中的作用及意义。方法利用异常黑胆质证造模法及二乙基亚硝胺诱导建立异常黑胆质型肝癌病证模型组、肝癌对照组,同时设立正常对照组。应用半定量PCR(RT-PCR)检测各组肝脏组织中STAT3基因的 mRNA水平的表达,免疫组织化学 SP法和蛋白免疫印记法(Western blot)检测各组肝脏组织中STAT3基因的蛋白水平的表达。结果肝癌对照组与正常对照组相比,STAT3基因的 mRNA水平明显上调,差异有统计学意义(P <0.01),异常黑胆质型肝癌病证模型组与肝癌对照组相比,STAT3基因的 mRNA水平明显上调,差异有统计学意义(P <0.01)。免疫组化 SP法及 Western blot结果显示肝癌对照组 STAT3蛋白表达水平高于正常对照组,差异有统计学意义(P <0.01),异常黑胆质型肝癌病证模型组 STAT3蛋白表达水平高于肝癌对照组,差异有统计学意义(P <0.05)。结论在异常黑胆质型肝癌的发生过程中,二乙基亚硝胺(DEN)是肝癌发生的主要原因,异常黑胆质体液起了促进作用。异常黑胆质体液可能通过对 STAT3肝癌基因的影响,促进异常黑胆质型肝癌的发生、发展。

목적:건립유오이의이상흑담질형간암병증대서모형,탐토신호전도급전록활화인자3(STAT3)기인재이상흑담질형간암병증발생발전중적작용급의의。방법이용이상흑담질증조모법급이을기아초알유도건립이상흑담질형간암병증모형조、간암대조조,동시설립정상대조조。응용반정량PCR(RT-PCR)검측각조간장조직중STAT3기인적 mRNA수평적표체,면역조직화학 SP법화단백면역인기법(Western blot)검측각조간장조직중STAT3기인적단백수평적표체。결과간암대조조여정상대조조상비,STAT3기인적 mRNA수평명현상조,차이유통계학의의(P <0.01),이상흑담질형간암병증모형조여간암대조조상비,STAT3기인적 mRNA수평명현상조,차이유통계학의의(P <0.01)。면역조화 SP법급 Western blot결과현시간암대조조 STAT3단백표체수평고우정상대조조,차이유통계학의의(P <0.01),이상흑담질형간암병증모형조 STAT3단백표체수평고우간암대조조,차이유통계학의의(P <0.05)。결론재이상흑담질형간암적발생과정중,이을기아초알(DEN)시간암발생적주요원인,이상흑담질체액기료촉진작용。이상흑담질체액가능통과대 STAT3간암기인적영향,촉진이상흑담질형간암적발생、발전。
Objective To establish abnormal savda disease carrying hepatocellular carcinoma animal model according to Uyghur traditional medicine.To investigate the important role and significance of STAT3 gene during the progress of originating and developing abnormal savda disease carrying Hepatocellular Car-cinoma.Methods Using methods of abnormal savda modeling and Diethylnirtosamine (DEN)inducing, we established abnormal savda disease carrying Hepatocellular Carcinoma model group,hepatocellular car-cinoma control group and normal control group.Applying Reverse transcriptase-polymerase chain reaction (RT-PCR),Immunohistochemistry dyeing and Western blotting were used to detect the mRNA and pro-tein expressions of STAT3 gene in abnormal savda disease carrying Hepatocellular Carcinoma.Results RT-PCR results showed that the STAT3 gene mRNA levels in hepatocellular carcinoma control group were significantly higher compared with normal control group.The difference is statistically significant (P<0.01).Immunohistochemistry dyeing results showed that hepatocellular carcinoma control group was significantly higher compared with normal control group.The difference was statistically significant (P <0.01).Abnormal savda disease carrying Hepatocellular Carcinoma model group was higher compared with hepatocellular carcinoma control group.The difference was statistically significant (P <0.05).Western-blot showed that hepatocellular carcinoma control group was higher compared with normal control group, which was statistically significant (P <0.05).Abnormal savda disease carrying Hepatocellular Carcinoma model group was higher compared with hepatocellular carcinoma control group,which was statistically significant (P <0.05 ).Conclusion DEN is the main hepatocellular carcinoma induction agent and abnormal savda hilit is the accelerative factor in the process of establishing abnormal savda disease carrying hepatocellular carcinoma model.The abnormal savda hilit may promote the occurrence and development of abnormal savda carrying hepatocellular carcinoma by influencing STAT3 gene of hepatocellular carcinoma.
