目的探讨邻苯二甲酸二(2-乙基己基)酯(DEHP)宫内暴露对子代雌性大鼠心脏的影响。方法自妊娠第1天直至分娩,SD妊娠大鼠分别予以灌胃玉米油、低剂量[10 mg/(kg·d)]DEHP、高剂量[100 mg/(kg·d)]DEHP。观察各组子代雌性大鼠一般状况(体重)、心肌组织学以及心脏的血流动力学指标。结果①与正常对照组比较,DEHP染毒组雌性仔鼠自出生至生后70 d的体重增长明显,差异有统计学意义(P<0.05)。于高剂量组的心率(HR)、左室舒张压( LVEDP)明显高于正常对照组及低剂量组,而左室收缩压( LVSP)、左室最大上升速率(+dp/dtmax )及左室最大下降速率(-dp/dtmax )明显低于正常对照组及低剂量组,差异有统计学意义( P<0.05)。③光镜下高剂量组子代雌性大鼠的心肌细胞肥大,心肌纤维排列紊乱,大量心肌细胞核核固缩、深染;电镜示大量心肌细胞核固缩、深染,心肌纤维排列紊乱、断裂,线粒体嵴排列大部分断裂、模糊。然而,正常对照组和低剂量组病理变化不明显。结论怀孕期间大剂量的DEHP接触可引起子代雌性大鼠体重增加、血流动力学异常和心肌细胞结构紊乱。

목적탐토린분이갑산이(2-을기기기)지(DEHP)궁내폭로대자대자성대서심장적영향。방법자임신제1천직지분면,SD임신대서분별여이관위옥미유、저제량[10 mg/(kg·d)]DEHP、고제량[100 mg/(kg·d)]DEHP。관찰각조자대자성대서일반상황(체중)、심기조직학이급심장적혈류동역학지표。결과①여정상대조조비교,DEHP염독조자성자서자출생지생후70 d적체중증장명현,차이유통계학의의(P<0.05)。우고제량조적심솔(HR)、좌실서장압( LVEDP)명현고우정상대조조급저제량조,이좌실수축압( LVSP)、좌실최대상승속솔(+dp/dtmax )급좌실최대하강속솔(-dp/dtmax )명현저우정상대조조급저제량조,차이유통계학의의( P<0.05)。③광경하고제량조자대자성대서적심기세포비대,심기섬유배렬문란,대량심기세포핵핵고축、심염;전경시대량심기세포핵고축、심염,심기섬유배렬문란、단렬,선립체척배렬대부분단렬、모호。연이,정상대조조화저제량조병리변화불명현。결론부잉기간대제량적DEHP접촉가인기자대자성대서체중증가、혈류동역학이상화심기세포결구문란。
Objective To investigate the cardiac effects of DEHP exposure during pregnancy on female rats off-spring. Methods Pregnant SD rats were gavaged from gestationa day 0 until birth with corn oil, low [ 10 mg/( kg·d) ] dose DEHP and high [100 mg/( kg·d) ] dose DEHP. Each group consists of two pregnant dams. The outcomes of general morphometry( weight) , cardiac histology and cardiac function were studied. Results ①When exposed to DEHP during pregnancy, body weight of female rats offspring increased from PND 0 (Postnatal day 0) to PND 70 . The increase level of weight was significantly higher in high dose group at PND 70 ( P <0.05 ) . ②Compared with control group and low dose DEHP group, the heart rate ( HR) and left ventricular end-diastolic pres-sure ( LVEDP) of female adult rats were deteriorated in the high dose DEHP group, while left ventricular systolic pressure ( LVSP) , maximal rise velocity of left ventricular pressure ( +dp/dtmax ) and maximal fall velocity of left ventricular pressure ( -dp/dtmax) of that were significantly decreased (P<0.05). ③The cardiac histology effects of high dose DEHP exposure as follows:Pathological observation demonstrated cardiac myocyte hypertrophy, myo-cardial fiber disarray, a large number of myocardial cells karyopycnosis and stained;electron microscopy showed a lot of myocardial cells karyopycnosis and stained, myocardial fiber disorder and fracture arrangement, most of mito-chondrial cristae were broken and blurred. However, no obvious change was found in the control group and low dose DEHP group. Conclusion High dose DEHP exposure in utero may lead weight increase, hemodynamic and myocardial abnormalities of female rats offspring.
