目的:研究逍遥散对氢溴酸东莨菪碱致阿尔茨海默病( AD )模型小鼠学习记忆能力及神经递质活性的影响。方法50只 SPF 级KM小鼠随机分为空白组、模型组、逍遥散低、中、高剂量组,腹腔注射氢溴酸东莨菪碱复制 AD小鼠模型并以逍遥散不同剂量进行干预防治,给药第6、8天水迷宫训练各组小鼠学习能力,第10天测定小鼠记忆能力,然后取脑组织测胆碱乙酰转移酶( ChAT)、乙酰胆碱酯酶( AchE)及单胺氧化酶( MAO)活性。结果逍遥散低、中、高剂量组均可提高小鼠学习记忆能力,降低潜伏时间,提高脑组织匀浆ChAT活性,降低AchE及MAO活性,但学习记忆能力及神经递质活性的改善皆以逍遥散高剂量组最为明显。结论逍遥散能较好改善AD模型小鼠学习记忆及神经递质活性,对 AD具有较好的防治作用。
目的:研究逍遙散對氫溴痠東莨菪堿緻阿爾茨海默病( AD )模型小鼠學習記憶能力及神經遞質活性的影響。方法50隻 SPF 級KM小鼠隨機分為空白組、模型組、逍遙散低、中、高劑量組,腹腔註射氫溴痠東莨菪堿複製 AD小鼠模型併以逍遙散不同劑量進行榦預防治,給藥第6、8天水迷宮訓練各組小鼠學習能力,第10天測定小鼠記憶能力,然後取腦組織測膽堿乙酰轉移酶( ChAT)、乙酰膽堿酯酶( AchE)及單胺氧化酶( MAO)活性。結果逍遙散低、中、高劑量組均可提高小鼠學習記憶能力,降低潛伏時間,提高腦組織勻漿ChAT活性,降低AchE及MAO活性,但學習記憶能力及神經遞質活性的改善皆以逍遙散高劑量組最為明顯。結論逍遙散能較好改善AD模型小鼠學習記憶及神經遞質活性,對 AD具有較好的防治作用。
목적:연구소요산대경추산동랑탕감치아이자해묵병( AD )모형소서학습기억능력급신경체질활성적영향。방법50지 SPF 급KM소서수궤분위공백조、모형조、소요산저、중、고제량조,복강주사경추산동랑탕감복제 AD소서모형병이소요산불동제량진행간예방치,급약제6、8천수미궁훈련각조소서학습능력,제10천측정소서기억능력,연후취뇌조직측담감을선전이매( ChAT)、을선담감지매( AchE)급단알양화매( MAO)활성。결과소요산저、중、고제량조균가제고소서학습기억능력,강저잠복시간,제고뇌조직균장ChAT활성,강저AchE급MAO활성,단학습기억능력급신경체질활성적개선개이소요산고제량조최위명현。결론소요산능교호개선AD모형소서학습기억급신경체질활성,대 AD구유교호적방치작용。
Objective To observe the effect of xiaoyao powder on the ability of learning and memory and neurotransmitter activity of Alzheimer′s disease ( AD) model mice made by scopolamine hydrobromide and investigate the effect and mechanism of xiaoyao powder trea -ting AD.Methods 50 SPF KM mice were randomly divided into blank control , model and low, middle, high dose xiaoyao powder groups . AD mice model was induced by scopolamine hydrobromide alkali intraperitoneal injection and prevented by different dosage of xiaoyao pow -der, brain tissue was measured after 1 h of injecting of scopolamine hydrobromide ,the levels of ChAT, AchE, MAO activity in brain tissue homogenate were detected .Results Different dosage of xiaoyao powder improved learning and memory ability and lower average latency time and improved ChAT activity and lower AchE and MAO content ,and high dose group of Xiaoyao powder was most obvious .Conclusions Xi-aoyao powder can better improve learning and memory ability and neurotransmitter activity of AD model mice and has good control effect on ADN.
