目的:探讨在皮肤慢性溃疡处注射异体骨髓间充质干细胞(BMSC)和自体富血小板血浆(PRP)对皮肤的再生和重建的作用。方法选择健康6~8周龄SD大鼠17只,取其中1只大鼠,采用全骨髓法体外分离培养BMSC,取传至第3代细胞,移植前48 h行DiI荧光标记。剩余16只大鼠麻醉后制作全层皮肤缺损模型,于其背部脊柱两侧制作4个全层皮肤缺损创面并随机分为4组。A组局部注射异体BMSC悬液联合PRP治疗,B组局部单纯注射异体BMSC悬液,C组局部单纯注射PRP治疗,D组局部注射等量生理盐水。治疗7、14 d采集标本观察结果。采用大体观察、苏木素-伊红染色、荧光显微镜观察创面愈合情况。结果体外分离培养的BMSC 具有成脂、成骨、成神经分化潜能。在治疗后7、14 d,A、B、C组创面愈合时间均短于D组,A组愈合时间比B、C组缩短。组织学观察发现A组新生真皮相应时间点的厚度、血管和胶原纤维均明显多于其他3组。结论在皮肤慢性溃疡处联合应用异体BMSC和自体PRP,能加速真皮的修复和重建。
目的:探討在皮膚慢性潰瘍處註射異體骨髓間充質榦細胞(BMSC)和自體富血小闆血漿(PRP)對皮膚的再生和重建的作用。方法選擇健康6~8週齡SD大鼠17隻,取其中1隻大鼠,採用全骨髓法體外分離培養BMSC,取傳至第3代細胞,移植前48 h行DiI熒光標記。剩餘16隻大鼠痳醉後製作全層皮膚缺損模型,于其揹部脊柱兩側製作4箇全層皮膚缺損創麵併隨機分為4組。A組跼部註射異體BMSC懸液聯閤PRP治療,B組跼部單純註射異體BMSC懸液,C組跼部單純註射PRP治療,D組跼部註射等量生理鹽水。治療7、14 d採集標本觀察結果。採用大體觀察、囌木素-伊紅染色、熒光顯微鏡觀察創麵愈閤情況。結果體外分離培養的BMSC 具有成脂、成骨、成神經分化潛能。在治療後7、14 d,A、B、C組創麵愈閤時間均短于D組,A組愈閤時間比B、C組縮短。組織學觀察髮現A組新生真皮相應時間點的厚度、血管和膠原纖維均明顯多于其他3組。結論在皮膚慢性潰瘍處聯閤應用異體BMSC和自體PRP,能加速真皮的脩複和重建。
목적:탐토재피부만성궤양처주사이체골수간충질간세포(BMSC)화자체부혈소판혈장(PRP)대피부적재생화중건적작용。방법선택건강6~8주령SD대서17지,취기중1지대서,채용전골수법체외분리배양BMSC,취전지제3대세포,이식전48 h행DiI형광표기。잉여16지대서마취후제작전층피부결손모형,우기배부척주량측제작4개전층피부결손창면병수궤분위4조。A조국부주사이체BMSC현액연합PRP치료,B조국부단순주사이체BMSC현액,C조국부단순주사PRP치료,D조국부주사등량생리염수。치료7、14 d채집표본관찰결과。채용대체관찰、소목소-이홍염색、형광현미경관찰창면유합정황。결과체외분리배양적BMSC 구유성지、성골、성신경분화잠능。재치료후7、14 d,A、B、C조창면유합시간균단우D조,A조유합시간비B、C조축단。조직학관찰발현A조신생진피상응시간점적후도、혈관화효원섬유균명현다우기타3조。결론재피부만성궤양처연합응용이체BMSC화자체PRP,능가속진피적수복화중건。
Objective To investigate the effects of local injection with exogenous bone marrow mes-enchymal stem cells (BMSCs)and autologous platelet-rich plasma on skin regeneration and reconstruction in rats with chronic skin ulcer. Methods Seventeen SD rats aged 6-8 weeks were used in this study. BMSCs were isolated from bone marrow of one rat. The 3rd passage of BMSCs was DiI-labeled 48 hours before trans-plantation. Full thickness skin defect model with four wounds on the back was established in the remaining 16 rats after anesthesia. Four wounds of the rats were randomly divided into 4 groups. Skins wounds were locally injected with BMSCs and PRP in group A,BMSCs in group B,PRP in group C,and physiological saline solu-tion in group D. Histological analysis was performed to evaluate the velocity and the quality of the wound. Fluo-rescence microscope was used to evaluate the wound healing 7 days and 14 days after treatment. Results BM-SCs were capable of adipogenic,osteogenic and chondrogenetic differentiation in vitro. On day 7 and day 14, wound healing times of group A,B,C were significantly shorter than that of group D. And wound healing time of group A was shorter than those of group B and C. Histological analysis showed that the depth of the renas-cence dermis,the number of the blood vessel and collagen fiberall in group A were all better than those in any other groups. Conclusions In chronic skin ulcers,combined local application of exogenous BMSCs and autol-ogous PRP can accelerate the wound repair and reconstruction of the dermis.
