为了研究饲粮添加L-精氨酸( Arg)或N-氨甲酰谷氨酸( NCG)对环江香猪肌肉脂肪酸含量和脂质代谢相关基因表达的影响,选用初始体重为(28.14±3.45) kg的环江香猪24头,随机分为3组,每组4个重复,每个重复2头猪。 Arg 组和 NCG 组分别在基础饲粮中添加0.83%L-Arg+0.88%米糠和0.08%NCG +1.71%L -丙氨酸,对照组在基础饲粮中添加1.71%L-丙氨酸。饲喂45 d后称取体重,放血处死,采集肝脏和背最长肌,测定背最长肌中脂肪酸的含量、肝脏和背最长肌中脂质代谢相关基因的表达。结果表明,与对照组相比,饲粮添加Arg可显著降低肌肉C18:0含量( P<0.05),显著下调肝脏乙酰CoA羧化酶α( ACCα)以及肌肉ACCα和过氧化物酶增生物激活受体γ( PPARγ) mRNA 表达( P <0.05),显著上调肝脏PPARγ mRNA表达(P<0.05);饲粮添加NCG可显著下调肝脏ACCα和脂蛋白脂酶(LPL)以及肌肉ACCα、脂肪酸合成酶( FAS )和 LPL mRNA 表达( P <0.05),显著上调肌肉 PPARγmRNA表达( P<0.05)。上述结果提示,饲粮添加Arg或NCG可调节肝脏和背最长肌中脂质代谢相关基因的表达,并降低背最长肌中C18:0的含量。

This study was conducted to determine the effects of dietary supplemented with L-arginine ( Arg) or N-carbamylglutamate ( NCG) on intramuscular contents of fatty acids and the gene expression levels related to lipid metabolism in Huanjiang mini-pigs. Tweety-four Huanjiang mini-pigs with initial body weight ( BW) of (28. 14±3. 45) kg were randomly assigned into 3 groups with 4 replicates in each group and 2 pigs per repli-cate, representing Arg, NCG and control groups, and pigs were supplemented with 0. 83% L-Arg + 0. 88%rice bran, 0. 08% NCG+1. 71% L-alanine, and 1. 71% L-alanine in a basal diet, respectively. After a 45-day feeding period, the liver and longissimus dorsi were obtained for determining intramuscular contents of fat-ty acids and the gene expression levels related to lipid metabolism. The results showed that compared with the control group, dietary supplemented with Arg significantly decreased muscle content of C18:0 (P<0. 05), as well as the expression levels of acetyl-CoA carboxylaseα( ACCα) mRNA in liver, and ACCαand peroxisome proliferator activated receptor-γ( PPARγ) mRNA in muscle ( P<0 . 05 ) , while significantly increased the ex-pression level of PPARγ mRNA in liver ( P<0 . 05 ); dietary supplementation of NCG significantly decreased the expression levels of ACCαand lipoprteinlipase ( LPL) mRNA in liver ( P<0 . 05 ) , as well as ACCα, fatty acid synthetase ( FAS) and LPL mRNA in muscle, while significantly increased the expression level of PPARγmRNA in muscle ( P<0 . 05 ) . These results indicate that dietary supplemented with Arg or NCG have a regu-lating effect on gene expression related to lipid metabolism in the liver and longissimus dorsi, and decrease the content of C18:0 in longissimus dorsi.
