Experimental cold syndrome of primary dysmenorrheal model rats%Deqi%μ-receptor of uterine tension
目的:观察不同针刺刺激量对寒凝证类痛经大鼠模型的镇痛效应及子宫组织μ受体表达的影响。方法:按随机数字表法将64只雌性SD大鼠,随机分为4组,每组16只,分别为:盐水组、寒凝证类痛经模型组、刺激量A组、刺激量B组。除盐水组外均给予大鼠注射苯甲酸雌二醇和缩宫素2U/只,并于第1天至第5天放入-25℃冰柜冷冻,制备寒凝证类痛经大鼠模型;盐水组每日给予同等剂量的生理盐水。第10天注射缩宫素后,刺激量A组和刺激量B组以不同刺激量进行针刺治疗。记录疼痛即刻开始20 min扭体评分和次数、子宫收缩波;以RealTime-PCR法测定子宫组织内μ受体。结果:①与盐水组比较,寒凝证类痛经模型组扭体潜伏期均明显缩短、扭体次数明显增多、扭体总分明显增多(P<0.01)。与寒凝证类痛经模型组比较,刺激量A组扭体潜伏期明显延长、扭体次数减少、扭体评分减少(P<0.01)。与刺激量A组比较,刺激量B组扭体潜伏期明显缩短、扭体次数明显增多、扭体评分明显升高(P<0.01);②与盐水组相比,寒凝证类痛经模型组子宫收缩波个数明显增加(P<0.01),子宫收缩波峰峰值显著升高(P<0.01),子宫活动度显著增加(P<0.01);与寒凝证类痛经模型组比较,刺激量A组显著减少了子宫收缩波个数(P<0.01)并降低了子宫活动度(P<0.05),并且在减少子宫收缩波个数方面优于刺激量B组(P<0.05);③与寒凝证类痛经模型组比较,刺激量A组子宫组织中μ受体表达显著增高刺激量A组(P<0.01),刺激量B组(P<0.05)。结论:刺激量A组可明显减轻寒凝证类痛经大鼠疼痛,缓解子宫痉挛性收缩,其与调节阿片受体的水平密切相关,提示刺激量A组对寒凝证类痛经大鼠具有明显的镇痛效应。

목적:관찰불동침자자격량대한응증류통경대서모형적진통효응급자궁조직μ수체표체적영향。방법:안수궤수자표법장64지자성SD대서,수궤분위4조,매조16지,분별위:염수조、한응증류통경모형조、자격량A조、자격량B조。제염수조외균급여대서주사분갑산자이순화축궁소2U/지,병우제1천지제5천방입-25℃빙거냉동,제비한응증류통경대서모형;염수조매일급여동등제량적생리염수。제10천주사축궁소후,자격량A조화자격량B조이불동자격량진행침자치료。기록동통즉각개시20 min뉴체평분화차수、자궁수축파;이RealTime-PCR법측정자궁조직내μ수체。결과:①여염수조비교,한응증류통경모형조뉴체잠복기균명현축단、뉴체차수명현증다、뉴체총분명현증다(P<0.01)。여한응증류통경모형조비교,자격량A조뉴체잠복기명현연장、뉴체차수감소、뉴체평분감소(P<0.01)。여자격량A조비교,자격량B조뉴체잠복기명현축단、뉴체차수명현증다、뉴체평분명현승고(P<0.01);②여염수조상비,한응증류통경모형조자궁수축파개수명현증가(P<0.01),자궁수축파봉봉치현저승고(P<0.01),자궁활동도현저증가(P<0.01);여한응증류통경모형조비교,자격량A조현저감소료자궁수축파개수(P<0.01)병강저료자궁활동도(P<0.05),병차재감소자궁수축파개수방면우우자격량B조(P<0.05);③여한응증류통경모형조비교,자격량A조자궁조직중μ수체표체현저증고자격량A조(P<0.01),자격량B조(P<0.05)。결론:자격량A조가명현감경한응증류통경대서동통,완해자궁경련성수축,기여조절아편수체적수평밀절상관,제시자격량A조대한응증류통경대서구유명현적진통효응。
Objective:To investigate the effect of different acupuncture stimulus on pains and expression of μopioid re-ceptor of primary dysmenorrheal model rats with cold syndrome .Methods:Female rats in anestrus were randomly divided into four groups , which included saline control group , cold syndrome of primary dysmenorrheal model control group (model control group), A stimuli group and B Stimuli group .Saline control group rats were injected 0.9%Nacl.All other rats were injected Estradiol Benzoate .From the first day to the fifth day rats ( except saline control group ) were put into -25℃freezer to make cold syndrome of primary dysmenorrheal model rats .Different amounts of acupuncture were used in A Stimuli group and B Stimuli group .BL-420 F logger of experimental system was applied to record times and intensity of uterine contraction in 20 minutes and μ-receptor in uterine was detected by RealTime -PCR.Results:①Model control group comparing with the saline control group: writhing latency significantly decreased (P<0.01), writhing number and writhing number significantly increased ( P<0 .01 ) .A Stimuli group comparing with the model con-trol group:writhing latency significantly increased ( P<0 .01 ) , writhing number and writhing number significantly de-creased ( P <0 .01 ) .B Stimuli group comparing with A Stimuli Group writhing latency significantly decreased ( P<0.01 );②writhing number and writhing number significantly increased ( P<0 .01 );Model control group comparing with the saline control group:the times ,peak to peak of uterine contraction and intensity of uterine contraction significantly increased ( P<0 .01 ) , which meant the smooth muscle of uterus was induced spasmodic contraction .A Stimuli group comparing with the model control group:the times and the intensity of uterine contraction of uterine were reduced signifi-cantly (P<0.05) and better than B Stimuli group (P<0.05).③A Stimuli group increased the amount of μopioid re-ceptor in uterus tissue(A Stimuli group,P<0.01;B Stimuli group,P<0.05).Conclusion:A Stimuli group can reduce spasmodic contraction of the smooth muscle of uterus significantly , which closely related to expression of opioid receptor . A Stimuli group indicates significantly analgesia on experimental cold syndrome of primary dysmenorrheal model rats .
