本试验旨在探讨饲料中添加α-酮戊二酸( AKG)对草鱼热休克蛋白70( HSP70)和过氧化物酶体增殖物激活受体γ(PPARγ)mRNA表达的影响。选取平均体重为(35.66±0.39) g的健康草鱼450尾,随机分为5组(每组3个重复,每个重复30尾鱼):对照组饲喂基础饲料,试验组分别饲喂以0.25%、0.50%、0.75%、1.00%AKG等量替换基础饲料中次粉的试验饲料。试验期为60 d,试验结束后采用实时荧光定量PCR方法检测草鱼肝胰脏、鳃、肾脏及脾脏中HSP70和PPARγmRNA的相对表达量。结果表明:HSP70和PPARγmRNA的相对表达量在不同组织中存在差异,二者均在肾脏中高度表达,HSP70 mRNA的相对表达量在肾脏、肝胰脏、脾脏、鳃中依次降低,而PPARγmRNA的相对表达量在肾脏、肝胰脏、鳃、脾脏中依次降低。饲料中添加AKG显著或极显著影响HSP70和PPARγmRNA在鳃、肾脏、脾脏中的表达以及PPARγmRNA在肝胰脏中的表达( P<0.05或 P<0.01)。与对照组相比,1.00%AKG组鳃中HSP70、PPARγmRNA的相对表达量显著或极显著增加( P<0.05或P<0.01),0.75%AKG组脾脏中HSP70、PPARγmRNA的相对表达量显著或极显著增加( P<0.05或P<0.01),各AKG添加组肾脏中HSP70、PPARγmRNA的相对表达量均显著降低( P<0.05);0.25%AKG组肝胰脏中PPARγmRNA的相对表达量极显著高于其他各组(P<0.01),但肝胰脏中HSP70 mRNA的相对表达量在各组间差异不显著( P>0.05)。综合以上结果,建议草鱼(体重在35 g左右)饲料中AKG的添加量为0.75%。

본시험지재탐토사료중첨가α-동무이산( AKG)대초어열휴극단백70( HSP70)화과양화물매체증식물격활수체γ(PPARγ)mRNA표체적영향。선취평균체중위(35.66±0.39) g적건강초어450미,수궤분위5조(매조3개중복,매개중복30미어):대조조사위기출사료,시험조분별사위이0.25%、0.50%、0.75%、1.00%AKG등량체환기출사료중차분적시험사료。시험기위60 d,시험결속후채용실시형광정량PCR방법검측초어간이장、새、신장급비장중HSP70화PPARγmRNA적상대표체량。결과표명:HSP70화PPARγmRNA적상대표체량재불동조직중존재차이,이자균재신장중고도표체,HSP70 mRNA적상대표체량재신장、간이장、비장、새중의차강저,이PPARγmRNA적상대표체량재신장、간이장、새、비장중의차강저。사료중첨가AKG현저혹겁현저영향HSP70화PPARγmRNA재새、신장、비장중적표체이급PPARγmRNA재간이장중적표체( P<0.05혹 P<0.01)。여대조조상비,1.00%AKG조새중HSP70、PPARγmRNA적상대표체량현저혹겁현저증가( P<0.05혹P<0.01),0.75%AKG조비장중HSP70、PPARγmRNA적상대표체량현저혹겁현저증가( P<0.05혹P<0.01),각AKG첨가조신장중HSP70、PPARγmRNA적상대표체량균현저강저( P<0.05);0.25%AKG조간이장중PPARγmRNA적상대표체량겁현저고우기타각조(P<0.01),단간이장중HSP70 mRNA적상대표체량재각조간차이불현저( P>0.05)。종합이상결과,건의초어(체중재35 g좌우)사료중AKG적첨가량위0.75%。
This experiment was conducted to investigate the effects of α-ketoglutarate ( AKG) on heat shock protein 70 ( HSP70 ) and peroxisome proliferator activated receptor γ( PPARγ) mRNA expressions of grass carp (Ctenopharynodon idellus).A total of 450 grass carp with an average body weight of (35.66±0.39) g were randomly allotted to 5 groups with 3 replicates per group and 30 fish per replicate.Fish in control group were fed a basal diet, while those in experimental groups were fed experiemental diets supplemented with 0.25%, 0.50%, 0.75%and 1.00%AKG to substitute equivalent wheat middling on the basis of basal diet, re-specitively.The experiment lasted for 60 days.An real-time quantitative PCR method was used to measure the relative expression levels of HSP70 and PPARγmRNA in hepatopancreas, gill, kidney and spleen of grass carp at the end of experiment.The results showed as follows:there were differences in the relative expression levels of HSP70 and PPARγmRNA among different tissues, and both of them were highly expressed in kindy. The relative expression level of HSP70 mRNA in kindy, hepatopancreas, spleen and gill was systematically de-creased, while the relative expression level of PPARγmRNA in kindy, hepatopancreas, gill and spleen was systematically decreased.AKG supplementation significantly affected the expressions of HSP70 and PPARγmRNA in gill, kindy and spleen and the expression of HSP70 mRNA in hepatopancreas ( P<0.05 or P<0.01) . Compared with the control group, the relative expression levels of HSP70 and PPARγmRNA in gill were sig-nificantly increased in the 1.00%AKG group ( P<0.05 or P<0.01), and the relative expression levels of HSP70 and PPARγmRNA in spleen were significantly increased in the 0.75%AKG group ( P<0.05 or P<0.01) , while the relative expression levels of HSP70 and PPARγmRNA in kindy were significantly decreased in all AKG supplemental groups ( P<0.05) .The relative expression level of PPARγmRNA in hepatopancreas in 0.25%AKG group was significantly higher than that in other groups ( P<0.01) , however, the relative ex-pression level of HSP70 mRNA in hepatopancreas showed no significant difference among all groups ( P>0.05) .Based on above results, the recommended supplemental level of AKG in diets of grass carp ( the body weight is about 35 g) is 0.75%.
