目的:探讨缺氧诱导因子-1α(HIF-1α)在磷酸二酯酶4抑制剂罗氟司特调节人气管上皮细胞黏蛋白 MUC5AC 表达中的作用。方法制备香烟烟雾提取物,分别用罗氟司特或香烟烟雾提取物或二者同时干预支气管上皮细胞16 HBE。采用 RT-PCR 法检测细胞中 MUC5AC 及 HIF-1α mRNA 表达量,Western blot 检测 MUC5AC 及 HIF-1α蛋白的表达情况。结果与对照组相比,香烟烟雾提取物能够从转录和蛋白水平显著促进支气管上皮细胞16 HBE 中 MUC5AC 及 HIF-1α的表达,而罗氟司特干预可以明显抑制香烟烟雾提取物诱导的 HIF-1α,并降低 MUC5AC 的表达。结论磷酸二酯酶4抑制剂罗氟司特可以通过降低 HIF-1α而抑制香烟烟雾提取物所诱导的支气管上皮细胞 MUC5AC 的表达。
目的:探討缺氧誘導因子-1α(HIF-1α)在燐痠二酯酶4抑製劑囉氟司特調節人氣管上皮細胞黏蛋白 MUC5AC 錶達中的作用。方法製備香煙煙霧提取物,分彆用囉氟司特或香煙煙霧提取物或二者同時榦預支氣管上皮細胞16 HBE。採用 RT-PCR 法檢測細胞中 MUC5AC 及 HIF-1α mRNA 錶達量,Western blot 檢測 MUC5AC 及 HIF-1α蛋白的錶達情況。結果與對照組相比,香煙煙霧提取物能夠從轉錄和蛋白水平顯著促進支氣管上皮細胞16 HBE 中 MUC5AC 及 HIF-1α的錶達,而囉氟司特榦預可以明顯抑製香煙煙霧提取物誘導的 HIF-1α,併降低 MUC5AC 的錶達。結論燐痠二酯酶4抑製劑囉氟司特可以通過降低 HIF-1α而抑製香煙煙霧提取物所誘導的支氣管上皮細胞 MUC5AC 的錶達。
목적:탐토결양유도인자-1α(HIF-1α)재린산이지매4억제제라불사특조절인기관상피세포점단백 MUC5AC 표체중적작용。방법제비향연연무제취물,분별용라불사특혹향연연무제취물혹이자동시간예지기관상피세포16 HBE。채용 RT-PCR 법검측세포중 MUC5AC 급 HIF-1α mRNA 표체량,Western blot 검측 MUC5AC 급 HIF-1α단백적표체정황。결과여대조조상비,향연연무제취물능구종전록화단백수평현저촉진지기관상피세포16 HBE 중 MUC5AC 급 HIF-1α적표체,이라불사특간예가이명현억제향연연무제취물유도적 HIF-1α,병강저 MUC5AC 적표체。결론린산이지매4억제제라불사특가이통과강저 HIF-1α이억제향연연무제취물소유도적지기관상피세포 MUC5AC 적표체。
Objective To investigate the role of HIF-1αin phosphodiesterase-4 inhibitor roflumilast mediated effect on cigarette smoke extract on induced MUC5AC expression in human bronchial epithelial. Methods Cigarette smoke extract was prepared.And human bronchial epithelial cell 1 6 HBE was treated with roflumilast or cigarette smoke extract or both.Then the MUC5AC and HIF-1α mRNA expression were measured by RT-PCR,the protein expression of MUC5AC and HIF-1α were detected by Western blot.Results As compared with the control group,the expression of MUC5AC and HIF-1α were significantly increased after cells treated with cigarette smoke extract at both mRNA and protein level. While,the cigarette smoke extract induced upregulation of MUC5AC and HIF-1α were inhibited by roflumilast intervention.Conclusions Phosphodiesterase-4 inhibitor roflumilast can reverse the cigarette smoke extract induced MUC5AC upregulation by inhibiting HIF-1αexpression.
