目的:探讨糜酶途径对犬血管平滑肌细胞( VSMC)的血管紧张素Ⅱ( AngⅡ)生成及细胞增殖的作用。方法通过组织块贴壁法培养犬动脉VSMC,分为4组:对照组、AngⅠ组、卡托普利组,糜酶抑素组。对照组中仅为空白培养液,其他各组均加入AngⅠ,卡托普利组和糜酶抑素组再分别加入相应的干预药物。应用台盼蓝染色法和MTT法检测各组VSMC增殖的变化,放射免疫法检测培养液中AngⅡ含量等。结果成功培养出犬VSMC,卡托普利组和糜酶抑素组细胞计数及吸光度值均显著低于AngⅠ组(均P<0.05),而且糜酶抑素组较卡托普利组更明显(P<0.05);糜酶抑素组AngⅡ含量较AngⅠ组和卡托普利组低(P<0.05)。结论糜酶在犬VSMC的AngⅡ生成及细胞增殖过程中起着重要作用,其抑制剂可抑制VSMC增殖。

목적:탐토미매도경대견혈관평활기세포( VSMC)적혈관긴장소Ⅱ( AngⅡ)생성급세포증식적작용。방법통과조직괴첩벽법배양견동맥VSMC,분위4조:대조조、AngⅠ조、잡탁보리조,미매억소조。대조조중부위공백배양액,기타각조균가입AngⅠ,잡탁보리조화미매억소조재분별가입상응적간예약물。응용태반람염색법화MTT법검측각조VSMC증식적변화,방사면역법검측배양액중AngⅡ함량등。결과성공배양출견VSMC,잡탁보리조화미매억소조세포계수급흡광도치균현저저우AngⅠ조(균P<0.05),이차미매억소조교잡탁보리조경명현(P<0.05);미매억소조AngⅡ함량교AngⅠ조화잡탁보리조저(P<0.05)。결론미매재견VSMC적AngⅡ생성급세포증식과정중기착중요작용,기억제제가억제VSMC증식。
Objective To investigate the effect of chymase on the cell proliferation and angiotensin Ⅱ( AngⅡ) generation of dog vascular smooth muscle cells .Methods The dog artery smooth muscle cells were cultured and divided into four groups:Control group, Angiotensin Ⅰ(Ang Ⅰ) group, Captopril group and Chymostatin group .Cell counting was performed by trypan blues staining , while the ratio of light density by MTT and Ang Ⅱconcentration were measured in the culture medium .Results The cell counts were significantly reduced in Captopril group and Chymostatin group com -pared with Ang Ⅰgroup (P<0.05).The cell proliferation of Chymostatin group was significantly inhibited (P<0.05). Moreover, the concentration of Ang Ⅱin Chymostatin group was reduced significantly (P<0.05).Conclusion Chymase may play an important role in the generation of Ang Ⅱand the proliferation of cultured dog vascular smooth muscle cells , via which chymostatin may inhibit cell proliferation .
