目的:观察过氧化物酶体增殖物激活受体-γ( PPAR-γ)的配体15-脱氧-△12,14-前列腺素J2(15d-PGJ2)对兔耳增生性瘢痕的影响,探讨15d-PGJ2防治增生性瘢痕的机制和可行性。方法选取新西兰大白兔9只,在兔耳腹侧面制作1 cm ×1 cm全层皮肤缺损创面,每耳4个,共计72个,建立兔耳增生性瘢痕动物模型,随机分为实验组和对照组,分别用15d-PGJ2及生理盐水行瘢痕内注射,1次/d,连续注射7 d。停止注射后第3、6、17天取材。应用免疫组织化学方法检测CD34、转化生长因子-β1( TGF-β1)的表达。结果兔耳创面愈合后均出现不同程度类似人增生性瘢痕组织块,与对照组比较,实验组瘢痕逐渐变平变软,颜色变浅;在各时间点实验组CD34和TGF-β1的表达均较低,与对照组比较差异有统计学意义( P<0.05)。结论 PPAR-γ的配体15d-PGJ2可减少瘢痕内CD34、TGF-β1的含量,引起瘢痕萎缩,有一定的防治瘢痕增生的作用。

목적:관찰과양화물매체증식물격활수체-γ( PPAR-γ)적배체15-탈양-△12,14-전렬선소J2(15d-PGJ2)대토이증생성반흔적영향,탐토15d-PGJ2방치증생성반흔적궤제화가행성。방법선취신서란대백토9지,재토이복측면제작1 cm ×1 cm전층피부결손창면,매이4개,공계72개,건립토이증생성반흔동물모형,수궤분위실험조화대조조,분별용15d-PGJ2급생리염수행반흔내주사,1차/d,련속주사7 d。정지주사후제3、6、17천취재。응용면역조직화학방법검측CD34、전화생장인자-β1( TGF-β1)적표체。결과토이창면유합후균출현불동정도유사인증생성반흔조직괴,여대조조비교,실험조반흔축점변평변연,안색변천;재각시간점실험조CD34화TGF-β1적표체균교저,여대조조비교차이유통계학의의( P<0.05)。결론 PPAR-γ적배체15d-PGJ2가감소반흔내CD34、TGF-β1적함량,인기반흔위축,유일정적방치반흔증생적작용。
Objective To investigate the effect of 15-deoxy-△12,14 -prostagliandxin J2 (15d-PGJ2) on the expression of CD34 and TGF-β1 on hypertrophic scar in rabbit ears , and the possibility of 15d-PGJ2 in treatment of hy-pertrophic scar .Methods Ear hypertrophic scar model was established in 9 New Zealand white rabbits .Four wounds with total skin loses (1 cm ×1 cm) on the ventral side on each ear were constructed in all 9 rabbits.The wounds were randomly divided into the 15d-PGJ2 treatment group and control group , in which 10 μL 15d-PGJ2 and normal saline, respectively, were injected into the ear scar once a day for 7 days.On the 3rd, 6th and 17th day after the injection, 12 scars of each group were harvested .The expression of CD34 and TGF -β1 was detected by immunohistochemical method . Results Excessive dermal scars on rabbit ear that were similar to human hypertrophic scar were observed in the both two groups.Compared with control group , smaller, softer, flatter and color lighter scars were observed in treatment group .The expression of CD34 and TGF-β1 in the 15d-PGJ2 group was significantly down -regulated as compared with that in control group at different time points (P<0.05).Conclusion The 15d-PGJ2, ligand of PPAR-γ, can reduce the ex-pression of CD34 and TGF-β1 of hypertrophic scar in the rabbit ears and plays an important role in the prevention and treatment of hypertrophic scar .
