目的:探讨法舒地尔对缺血/再灌注损伤大鼠离体心脏细胞超微结构的保护作用。方法将SD大鼠25只随机分为对照组、I/R组、I/R+FL 组和I/R+FH 组。用改良的Langendorff灌流装置,K-H液灌流,I/R组预灌流20 min,停灌45 min,再灌30 min;I/R+FL组及I/R+FH 组于再灌注时分别在灌流液中加入盐酸法舒地尔注射液10 mg/kg和30 mg/kg;对照组连续灌流95 min。检测心肌组织超微结构及RhoA、ROCK-2蛋白表达。结果电镜显示,I/R组普遍出现收缩带,大量线粒体集聚、排列紊乱,单个线粒体肿胀、大小不一、嵴断裂、形成空泡,肌质网扩张,并出现细胞凋亡形态改变。 I/R+FL 组和I/R-FH 组心肌肌丝挛缩明显减轻,线粒体排列整齐,单个线粒体嵴排列有序、肿胀及空泡化明显减轻,仍有细胞凋亡形态改变。 RhoA、ROCK-2蛋白表达水平在I/R组及I/R+FL 组、I/R+FH 组明显高于对照组。结论法舒地尔对缺血/再灌注诱导的心肌超微结构损伤有保护作用。

목적:탐토법서지이대결혈/재관주손상대서리체심장세포초미결구적보호작용。방법장SD대서25지수궤분위대조조、I/R조、I/R+FL 조화I/R+FH 조。용개량적Langendorff관류장치,K-H액관류,I/R조예관류20 min,정관45 min,재관30 min;I/R+FL조급I/R+FH 조우재관주시분별재관류액중가입염산법서지이주사액10 mg/kg화30 mg/kg;대조조련속관류95 min。검측심기조직초미결구급RhoA、ROCK-2단백표체。결과전경현시,I/R조보편출현수축대,대량선립체집취、배렬문란,단개선립체종창、대소불일、척단렬、형성공포,기질망확장,병출현세포조망형태개변。 I/R+FL 조화I/R-FH 조심기기사련축명현감경,선립체배렬정제,단개선립체척배렬유서、종창급공포화명현감경,잉유세포조망형태개변。 RhoA、ROCK-2단백표체수평재I/R조급I/R+FL 조、I/R+FH 조명현고우대조조。결론법서지이대결혈/재관주유도적심기초미결구손상유보호작용。
Objective To investigate the protective effect of fasudial on the myocardial ultra -structure induced by ischemia-reperfusion (I/R) injury in rats.Methods Twenty-five Sprague Dawley rats were randomly divided into 4 groups:control group, I/R group, I/R+FL group and I/R+FH group.Using the Langendorff model in isolated working rat heart, after pre-perfusion 20 min, hearts from I/R group were subjected to 45 min of global ischemia followed by 30 min reperfusion.The hearts from I/R+FL group and I/R+FH group were given with perfusate fasudil hydrochloride injec-tion by 10 and 30 mg/kg, respectively, at reperfusion;while 95 min continuous perfusion was given to control hearts .The myocardial ultra-structure and the RhoA , ROCK-2 protein expression were analyzed by transmission electron microsco-py and confocal laser scanning electron microscope , respectively .Results According to electron microscopy analysis , contraction band , mitochondria gathering , arranging in disorder , swelling in individual , different in sizes , with cristae and vacuoles forming, sarcoplasmic reticulum dilatation, and apoptotic morphological changes were observed in I /R group.In I/R+F groups, the damage of myocardial myofilament and mitochondria were significantly reduced , though presenting with cell apoptosis morphological changes .RhoA and ROCK-2 protein expression levels were significantly higher in I/R group and I/R+F group than that in the control group .Conclusion Fasudil has a protective effect on the myocardial ul-trastructure changes induced by ischemia -reperfusion injury .
