目的:观察小蘖碱对氧化型低密度脂蛋白(oxLDL)诱导的大鼠胸主动脉平滑肌细胞(VSMCs)泡沫化及凝集素样氧化型低密度脂蛋白受体1(LOX-1)、核转录因子 P65(NF-κB P65)表达的影响。方法:将一部分体外培养的VSMCs分为空白对照组,泡沫化模型组(简称模型组),50μM小蘖碱对照组和 50μM小蘖碱干预组,油红 O染色后,观察各组VSMCs的泡沫化程度。另一部分体外培养的VSMCs同上分为空白对照组、模型组以及25μM小蘖碱干预组和50μM小蘖碱干预组,预刺激后Western-Blotting检测各组 VSMCs的 LOX-1蛋白和 NF-κB P65蛋白表达水平。结果:空白对照组和小蘖碱组VSMCs未泡沫化,LOX-1和NF-κB P65微量表达。模型组VSMCs中的脂滴聚集成团,形成泡沫化,LOX-1及NF-κB P65的表达较对照组明显增加(P<0.01),小蘖碱干预组泡沫化明显减轻,两小蘖碱干预组VSMCs LOX-1及NF-κB P65蛋白表达较模型组显著降低(P<0.01),高效量组作用更强(P<0.05,P<0.01)。结论:小檗碱可减轻平滑肌细胞源性泡沫细胞化程度,其作用机制可能与抑制 NF-κB和 LOX-1蛋白表达有关。

목적:관찰소얼감대양화형저밀도지단백(oxLDL)유도적대서흉주동맥평활기세포(VSMCs)포말화급응집소양양화형저밀도지단백수체1(LOX-1)、핵전록인자 P65(NF-κB P65)표체적영향。방법:장일부분체외배양적VSMCs분위공백대조조,포말화모형조(간칭모형조),50μM소얼감대조조화 50μM소얼감간예조,유홍 O염색후,관찰각조VSMCs적포말화정도。령일부분체외배양적VSMCs동상분위공백대조조、모형조이급25μM소얼감간예조화50μM소얼감간예조,예자격후Western-Blotting검측각조 VSMCs적 LOX-1단백화 NF-κB P65단백표체수평。결과:공백대조조화소얼감조VSMCs미포말화,LOX-1화NF-κB P65미량표체。모형조VSMCs중적지적취집성단,형성포말화,LOX-1급NF-κB P65적표체교대조조명현증가(P<0.01),소얼감간예조포말화명현감경,량소얼감간예조VSMCs LOX-1급NF-κB P65단백표체교모형조현저강저(P<0.01),고효량조작용경강(P<0.05,P<0.01)。결론:소벽감가감경평활기세포원성포말세포화정도,기작용궤제가능여억제 NF-κB화 LOX-1단백표체유관。
Objective:To investigate the effect of berberine on oxLDL induced foam cell formation and lectin-like oxidized low density lipoprotein receptor1(LOX-1),NF-κB expression in vascular smooth muscle Cells(VSMCs).Method:VSMC were cultured and classified into four groups:control group,foam cell model group(treated with oxLDL for 48h to establish the foam cell models),or 25uM berberine group and 50uM berberine group(different dose of ber-berine was pretreated for 2 hours).LOX-1 protein expression was detected by western-blot after treated with ox-LDL for 48h.Western-blot was applied to detect nuclear expression of NF-κB P65 protein after treated with oxLDL for 12h.Results:Compared with the foam cell models,oxLDL agregation and lipid droplets of foam cells in berberine treatment decreased significantly.The LOX-1 protein expression decreased at a low ebb after berberine treatment(P<0.05).On the other hand,with the increasing concentration of berberine treatment,the LOX-1 protein expres-sion decreased progressively (P<0.01).With two dosages of berberine,significantly difference was shown between berberine groups and oxLDL group for nuclear NF-κB P65 protein expression (P<0.01).Conclusion:Berberine in-hibits LOX-1 and nuclear NF-κB P65 protein expression in a dose-dependent manner.This may be part of the mech-anism of its anti-atherosclerotic effect.
