目的：构建 miRNA-491-5p 慢病毒过表达载体，筛选 miRNA-491-5p 稳定表达细胞株。方法：构建PCDH-511B-miRNA-491-5p载体，转染HEK293T细胞，通过实时荧光定量PCR检测miRNA-491-5p的含量。结果：实时荧光定量PCR检测显示转染慢病毒载体组miRNA-491-59表达水平是对照载体组的1000倍，证明目的基因已在转染细胞中成功表达。结论：成功构建了miRNA-491-5p慢病毒过表达载体和筛选出了miRNA-491-5p稳定表达细胞株。
目的：構建 miRNA-491-5p 慢病毒過錶達載體，篩選 miRNA-491-5p 穩定錶達細胞株。方法：構建PCDH-511B-miRNA-491-5p載體，轉染HEK293T細胞，通過實時熒光定量PCR檢測miRNA-491-5p的含量。結果：實時熒光定量PCR檢測顯示轉染慢病毒載體組miRNA-491-59錶達水平是對照載體組的1000倍，證明目的基因已在轉染細胞中成功錶達。結論：成功構建瞭miRNA-491-5p慢病毒過錶達載體和篩選齣瞭miRNA-491-5p穩定錶達細胞株。
목적：구건 miRNA-491-5p 만병독과표체재체，사선 miRNA-491-5p 은정표체세포주。방법：구건PCDH-511B-miRNA-491-5p재체，전염HEK293T세포，통과실시형광정량PCR검측miRNA-491-5p적함량。결과：실시형광정량PCR검측현시전염만병독재체조miRNA-491-59표체수평시대조재체조적1000배，증명목적기인이재전염세포중성공표체。결론：성공구건료miRNA-491-5p만병독과표체재체화사선출료miRNA-491-5p은정표체세포주。
Objective: To con struct the lentivirus expression vector of miRNA-491-5p and to screen cell lines for stable expression of miRNA-491-5p. Methods: The constructed recombinant vector was transfected to HEK293T cell. The miRNA-491-5p expression was detected by using real-time quantitative polymerase chain reaction ( PCR ) . Results: Real-time quantitative polymerase chain reaction showed that the over-expression of miRNA-491-5p of virus vector was 1 000 times of that of control vector group, which indicated that goal gene was expressed in transfected cells successfully. Conclusion: The over-expression virus vector of miRNA-491-5p have been successfully constructed and a cell lines for stable expression of miRNA-491-5p have been successfully screened.
