目的:观察决明子蒽醌苷对两肾一夹(2K1C)高血压大鼠的降压及肾损伤保护作用。方法复制2K1C高血压大鼠动物模型,随机分为模型组和决明子蒽醌苷高(0.4 g/kg)、低(0.2 g/kg)剂量组。另取10只大鼠作为假手术组。各组大鼠连续日一次灌胃给药6周,每周末检测大鼠尾动脉收缩压。末次给药后次日颈椎脱臼处死大鼠,肾组织苏木素-伊红及弹力纤维染色切片,显微镜下测量小叶间动脉、入球小动脉中膜厚度与管腔直径比值。计数高倍镜下每视野肾小管蛋白质管型数、肾间质淋巴细胞数,计算全片的均值。结果决明子蒽醌苷能明显降低2 K1 C高血压大鼠尾动脉收缩压( P <0.01或0.05)。决明子蒽醌苷能减缓2K1C高血压大鼠肾内小动脉重塑,小叶间动脉、入球小动脉中膜厚度与管腔直径比值较模型组明显降低( P <0.01或0.05)。决明子蒽醌苷能明显减弱2K1C高血压大鼠肾脏损伤程度,每高倍镜视野蛋白管型数及肾间质淋巴细胞数较模型组均明显减少( P <0.01或0.05);决明子蒽醌苷高剂量组降压及肾保护作用较低剂量组明显( P <0.01或0.05)。结论决明子蒽醌苷能明显降低2K1C高血压大鼠尾动脉收缩压且对高血压大鼠肾脏具有保护作用。

목적:관찰결명자은곤감대량신일협(2K1C)고혈압대서적강압급신손상보호작용。방법복제2K1C고혈압대서동물모형,수궤분위모형조화결명자은곤감고(0.4 g/kg)、저(0.2 g/kg)제량조。령취10지대서작위가수술조。각조대서련속일일차관위급약6주,매주말검측대서미동맥수축압。말차급약후차일경추탈구처사대서,신조직소목소-이홍급탄력섬유염색절편,현미경하측량소협간동맥、입구소동맥중막후도여관강직경비치。계수고배경하매시야신소관단백질관형수、신간질림파세포수,계산전편적균치。결과결명자은곤감능명현강저2 K1 C고혈압대서미동맥수축압( P <0.01혹0.05)。결명자은곤감능감완2K1C고혈압대서신내소동맥중소,소협간동맥、입구소동맥중막후도여관강직경비치교모형조명현강저( P <0.01혹0.05)。결명자은곤감능명현감약2K1C고혈압대서신장손상정도,매고배경시야단백관형수급신간질림파세포수교모형조균명현감소( P <0.01혹0.05);결명자은곤감고제량조강압급신보호작용교저제량조명현( P <0.01혹0.05)。결론결명자은곤감능명현강저2K1C고혈압대서미동맥수축압차대고혈압대서신장구유보호작용。
Objective To observe the effects of semen cassiae anthraquinone glycoside( SCAG)on hypertension and kidney injury pro-tection in two-kidney one clip(2K1C)hypertensive rats. Methods 2K1C hypertensive rats were randomly divided into model group,SCAG group(0. 2 g/kg and 0. 4 g/kg,respectively),and sham operation group with 10 rats in each group. The medicine was given for six weeks by in-tragastric administration. Tail artery systolic blood pressure was measured at every weekend. Rats were executed by the method of cervical vertebra dislocation. Specimens from kidney tissues were sliced and stained( HE and elastic fibers staining). The middle membrane thickness and the ves-sel lumen diameter of interlobular artery and afferent arteriole were measured and the ratio of thickness/lumen diameter was calculated. The number of protein cast and lymphocytes were counted in each field of vision at high magnification and the average was calculated. Results Compared with the model group,lower systolic blood pressure( P <0. 01 or 0. 05),and ratio of thickness/lumen diameter( P <0. 01 or 0. 05)were observed in SCAG group;and glomerular damage and small artery remodeling were ameliorated in SCAG group. In addition,the number of protein cast and lymphocytes were lower than the model group( P <0. 01 or 0. 05). 0. 4 g/kg of SCAG was more efficient in lowering blood pressure and kidney injury protection than 0. 2 g/kg of SCAG( P <0. 01 or 0. 05). Conclusion SCAG can significantly reduce tail artery systolic blood pressure in 2K1C hypertensive rats and ameliorate hypertension induced renal damage.
