本文旨在研究酸模叶蓼类黄酮的提取、纯化工艺。试验采用乙醇溶剂浸提,结合超声波辅助提取酸模叶蓼中的类黄酮。在单因素试验的基础上对提取剂乙醇浓度、提取温度、提取时间和料液比进行四因素三水平正交试验,得出酸模叶蓼类黄酮最佳提取条件为乙醇浓度80%,提取温度50℃,提取时间30 min,料液比1:20,在此最佳条件下酸模叶蓼类黄酮的得率可达到10%,粗提物纯度为37%。通过静态吸附、静态解吸及静态吸附动力学试验,确定采用NKA-9大孔树脂对酸模叶蓼类黄酮进行纯化。较好的动态吸附条件为上样液浓度9~12 mg/mL,上样液pH 4~5,径高比l:10,流速1.25 mL/min,采用70%乙醇以1.25 mL/min的流速洗脱,在此条件下得到的酸模叶蓼类黄酮纯度可达71%,较粗提物提高近1倍。

The present research studied the technoiogy of preparing and purified of flavonoids from Polygonum lapathifolium. With alcohol used as extraction solvent, flavonoids was extracted from leguminous Polygonum lapathifolium grazing by ultrasonic method. The final extracting condi-tions were optimized by the orthogonal experiment considering of extraction solvent, temperature of extraction, time of extraction and ratio of solid to liquid of extraction.. The yield of crude flavonoids from Polygonum lapathifolium is 10%, and content of crude flavonoids is 37% with the op-timum conditions: 80% alcohol, temperature 50℃, time 30 minutes, the ration of alcohol to materi-al 1:20. The macroporous absorption resin NKA-9 was selected as exclusive separation resin of fla-vonoids of Polygonum lapathifolium. The results shows: the better dynamic adsorption condition of Polygonum lapathifolium are concentration of sample 9-12mg/mL, pH 4-5, ration of diameter to height 1:10, the absorption velocity is limited in 1.25mL/min. With the conditions of the elution solvent is 70% ethanol under room temperature, the elution r velocity is 1.25mL/min. Under the con-dition the purity of Polygonum lapathifolium flavonoids up to 71%, 1 times of crude flavonoids.
