本试验旨在研究植物甾醇对去卵巢小鼠血清激素及乳腺组织中孕激素受体的影响。50只雌性小鼠,随机选40只进行去卵巢手术,恢复7 d。分组如下:正常组、去卵巢对照组、植物甾醇低、中、高3个剂量组(每日灌胃20、80和320 mg/kg植物甾醇)。连续灌胃3周,第22天采集血样和乳腺组织,对小鼠血清中的雌二醇、孕酮和催乳素水平进行检测,对乳腺组织的孕激素受体基因表达进行荧光定量检测。结果表明,去卵巢对照组小鼠雌二醇水平显著低于正常组(P<0.05)。植物甾醇组小鼠血清雌二醇水平持续升高,其中,高剂量组小鼠血清雌二醇水平与正常组最接近;与去卵巢对照组相比,去卵巢植物甾醇组小鼠血清孕酮水平均有提高,但差异不显著(P>0.05);与去卵巢对照组相比,去卵巢植物甾醇组小鼠催乳素水平先下降后上升,但他们之间无显著差异(P>0.05)。与去卵巢对照组相比,植物甾醇各剂量组小鼠孕激素受体基因相对表达量呈上升趋势,低、中、高剂量组小鼠孕激素受体基因相对表达量分别是去卵巢对照组的1.05、1.13和2.78倍。植物甾醇可改善去卵巢小鼠内源激素不平衡的状况,促进乳腺组织PR基因的表达。

본시험지재연구식물치순대거란소소서혈청격소급유선조직중잉격소수체적영향。50지자성소서,수궤선40지진행거란소수술,회복7 d。분조여하:정상조、거란소대조조、식물치순저、중、고3개제량조(매일관위20、80화320 mg/kg식물치순)。련속관위3주,제22천채집혈양화유선조직,대소서혈청중적자이순、잉동화최유소수평진행검측,대유선조직적잉격소수체기인표체진행형광정량검측。결과표명,거란소대조조소서자이순수평현저저우정상조(P<0.05)。식물치순조소서혈청자이순수평지속승고,기중,고제량조소서혈청자이순수평여정상조최접근;여거란소대조조상비,거란소식물치순조소서혈청잉동수평균유제고,단차이불현저(P>0.05);여거란소대조조상비,거란소식물치순조소서최유소수평선하강후상승,단타문지간무현저차이(P>0.05)。여거란소대조조상비,식물치순각제량조소서잉격소수체기인상대표체량정상승추세,저、중、고제량조소서잉격소수체기인상대표체량분별시거란소대조조적1.05、1.13화2.78배。식물치순가개선거란소소서내원격소불평형적상황,촉진유선조직PR기인적표체。
Experiment was conducted to study the effect of Phytosterols on the serum hormone and progesterone receptor in ovariectomized KM mice. 40 female mice was randomly selected from 50 for ovarian surgery, and was postoperative recovered for 7 days. The mice was grouped as follows: nor-mal, ovariectomized control group, phytosterols low, medium and high dose groups (daily fed with 20mg/kg, 80mg/kg, 320mg/kg phytosterols respectively). Mice were intragastric administered contin-uous for 3 weeks, blood samples and the breast tissue were collected on the 22nd day, the levels of serum estradiol, progesterone, prolactin were detected, fluorescence quantitative detection for progesterone receptor gene expression in breast tissue was conducted.The results showed that: Es-tradiol level in ovariectomized control group was significantly lower than the normal group (P<0.05). Serum estradiol level of phytosterols continue to rise, among them, serum estradiol levels of high dose group mice was the most closet to the normal group; Compared with the ovariectomized control group, serum progesterone levels in phytosterols ovariectomized mice were increased, but the difference was not significant (P>0.05); Compared with the ovariectomized control group, se-rum prolactin levels in phytosterols ovariectomized mice rised after the first decrease, but there was no significant difference between them (P>0.05);Compared with the ovariectomized control group, progesterone receptor gene relative expression level of ovariectomized mice in each dose group raised, the progesterone receptor gene relative expression levels of low, medium and high dose groups were 1.05 times, 1.13 times and 2.78 times than the ovariectomized control group. The results indicated that phytosterols can improve the imbalance of the endogenous hormones in ovari-ectomized mice, promote the expression of PR genes in breast tissue.
