目的:探讨维A酸对人A375黑素瘤株细胞黑素含量、酪氨酸酶活性及蛋白表达的影响。方法将培养的A375细胞分为维A酸组、8-甲氧补骨脂素(8-MOP)组、氢醌组、阴性对照组(A375细胞只加入相应溶媒,不加入药物)分别培养,于24 h、48 h和72 h测定黑素含量及酪氨酸酶活性;培养24 h Western检测酪氨酸酶蛋白的变化。结果A375细胞加入维A酸24 h、48 h、72 h后黑素含量及酪氨酸酶活性均较阴性对照组下降(P<0.05)。阴性对照组A375细胞酪氨酸酶蛋白含量测定值为(0.89±0.085),维A酸组A375细胞酪氨酸酶蛋白含量为(0.56±0.044)(t=3.811,P<0.05)。结论维A酸能抑制人A375黑素瘤株细胞黑素含量、酪氨酸酶活性及蛋白表达。

목적:탐토유A산대인A375흑소류주세포흑소함량、락안산매활성급단백표체적영향。방법장배양적A375세포분위유A산조、8-갑양보골지소(8-MOP)조、경곤조、음성대조조(A375세포지가입상응용매,불가입약물)분별배양,우24 h、48 h화72 h측정흑소함량급락안산매활성;배양24 h Western검측락안산매단백적변화。결과A375세포가입유A산24 h、48 h、72 h후흑소함량급락안산매활성균교음성대조조하강(P<0.05)。음성대조조A375세포락안산매단백함량측정치위(0.89±0.085),유A산조A375세포락안산매단백함량위(0.56±0.044)(t=3.811,P<0.05)。결론유A산능억제인A375흑소류주세포흑소함량、락안산매활성급단백표체。
ObjectiveTo evaluate the effects of retinoic acid (RA) on melanin content, activity and protein expression of tyrosinase in A375 cells.MethodsCultured A375 cells were classiifed into 4 groups: RA group, 8-methoxypsoralen (8-MOP) group, hydroquinone group, and negative control group received no treatment. Melanin content and tyrosinase activity were determined by NaOH solubilization assay and dopa-oxidation assay respectively at 24, 48 and 72 hours after the addition of RA into the medium. Western blot was performed to detect the protein expression of tyrosinase in A375 cells after 24 hours culture with RA.ResultsThe melanin content and tyrosinase activity decreased in A375 cells after being treated with RA for 24, 48 and 72 hours (P<0.05). The protein (gray scale) in A375 cells at 24 hours expression level of tyrosinase in negative control group was 0.89 ± 0.085, decreased to 0.56 ± 0.044 (t=3.811,P<0.05) after additional treatment with RA.ConclusionRA down-regulates activity of tyrosinase, inhibits protein expression of tyrosinase in A375 cells, as well as the biosynthesis of melanin.
