目的:观察人参皂苷对失神经大鼠腓肠肌MAFbx表达的影响,并探讨其延缓失神经腓肠肌萎缩的作用机制。方法:选用雌性 Wistar大鼠72只,随机分为3组:假失神经组(A)24只,失神经组(B)24只,人参皂苷治疗组(C)24只。于造模2、7、14、28 d后,用 Real time RT-PCR 和 Western blot 方法检测各组大鼠右侧腓肠肌MAFbx mRNA和蛋白的表达。结果:失神经组MAFbx表达在各时间点均明显高于假失神经组(P<0.01),且在失神经2 d达到峰值,而缓慢下降,失神经28 d 基本降至正常水平;人参皂苷组与失神经组各时间点相比,MAFbx 表达均明显降低(P <0.01)。结论:人参皂苷能通过减少MAFbx的表达而延缓失神经肌萎缩的发生。

목적:관찰인삼조감대실신경대서비장기MAFbx표체적영향,병탐토기연완실신경비장기위축적작용궤제。방법:선용자성 Wistar대서72지,수궤분위3조:가실신경조(A)24지,실신경조(B)24지,인삼조감치료조(C)24지。우조모2、7、14、28 d후,용 Real time RT-PCR 화 Western blot 방법검측각조대서우측비장기MAFbx mRNA화단백적표체。결과:실신경조MAFbx표체재각시간점균명현고우가실신경조(P<0.01),차재실신경2 d체도봉치,이완만하강,실신경28 d 기본강지정상수평;인삼조감조여실신경조각시간점상비,MAFbx 표체균명현강저(P <0.01)。결론:인삼조감능통과감소MAFbx적표체이연완실신경기위축적발생。
Obj ective:To investigate the effect and mechanism of Renshenzaogan on MAFBx expression in a rat model of denervated skeletal muscle atrophy.Methods:Seventy-two adult female Wistar rats were randomly divided into three groups:false nerve group (group A,n=24),the denervated gastrocnemius model group (group B,n=24),Renshenzaogan treatment group (group C,n=24).The expression of MAFbx mRNA and protein was detected by Real-time-PCR and Western blot,respectively.Results:The expression of MAFbx in the denervated group was significantly higher than that of false nerve group at each time point(P<0.01).It reached a peak at 2 days after the denervation,and then it declined slowly and returned the normal level after 28 days.The level of MAFbx in Renshenzaogan treatment group was significantly lower than those of the denervated group at each time point(P<0.01).Conclusion:Renshenzaogan can ameliorate skeletal muscle atrophy by decreasing the expression of MAFbx.
