目的:研究在孕期及哺乳期肥胖母鼠进行槲皮素干预后,对其F1代仔鼠体重和胰岛素样生长因子-1(insulin-like growth factors-1, IGF-1)mRNA表达的影响。方法:健康雌性SD大鼠断乳后,随机抽取8只喂养基础饲料作为空白对照组,其余喂养高脂饲料。当高脂饲料组大鼠平均体重超过空白对照组大鼠平均体重20%时,认为造模成功。将造模成功后的母鼠与健康雄鼠交配受孕,肥胖孕鼠随机分成4组,每组8只,整个孕期和哺乳期喂食高脂饲料,并分别以0 mg/kg体重(高脂对照组)、50 mg/kg体重、100 mg/kg体重、200 mg/kg体重剂量的槲皮素灌胃;空白对照组整个实验期喂食基础饲料并以溶剂灌胃。各组孕鼠自然分娩,记录F1代仔鼠出生体重及生后d7, d21,d56和d98体重,并检测F1代仔鼠肝组织IGF-1 mRNA的表达水平。结果:高脂对照组F1代仔鼠出生体重显著高于空白对照组。随槲皮素干预剂量增大,高脂对照组F1代仔鼠出生体重逐渐降低,接近空白对照组水平。槲皮素在一定程度上抑制F1代仔鼠肝组织IGF-1 mRNA的表达,持续到成年期,200 mg/kg体重效果最显著。结论:肥胖孕鼠F1代仔鼠出生体重显著增加,而孕期槲皮素干预可有效逆转F1代仔鼠体重,并控制F1代仔鼠断乳前体重过快增长。200 mg/kg体重的槲皮素能够有效抑制F1代仔鼠IGF-1 mRNA的表达,从而抑制细胞过度增殖和分化。

목적:연구재잉기급포유기비반모서진행곡피소간예후,대기F1대자서체중화이도소양생장인자-1(insulin-like growth factors-1, IGF-1)mRNA표체적영향。방법:건강자성SD대서단유후,수궤추취8지위양기출사료작위공백대조조,기여위양고지사료。당고지사료조대서평균체중초과공백대조조대서평균체중20%시,인위조모성공。장조모성공후적모서여건강웅서교배수잉,비반잉서수궤분성4조,매조8지,정개잉기화포유기위식고지사료,병분별이0 mg/kg체중(고지대조조)、50 mg/kg체중、100 mg/kg체중、200 mg/kg체중제량적곡피소관위;공백대조조정개실험기위식기출사료병이용제관위。각조잉서자연분면,기록F1대자서출생체중급생후d7, d21,d56화d98체중,병검측F1대자서간조직IGF-1 mRNA적표체수평。결과:고지대조조F1대자서출생체중현저고우공백대조조。수곡피소간예제량증대,고지대조조F1대자서출생체중축점강저,접근공백대조조수평。곡피소재일정정도상억제F1대자서간조직IGF-1 mRNA적표체,지속도성년기,200 mg/kg체중효과최현저。결론:비반잉서F1대자서출생체중현저증가,이잉기곡피소간예가유효역전F1대자서체중,병공제F1대자서단유전체중과쾌증장。200 mg/kg체중적곡피소능구유효억제F1대자서IGF-1 mRNA적표체,종이억제세포과도증식화분화。
Objective:To study the effect of quercetin given during pregnancy and lactation period of obese rats on weight and expression of insulin-like growth factors-1 ( IGF-1 ) mRNA of the offspring . Methods:In the study , 8 healthy weaning female SD rats were randomly selected to feed the basal diet and as blank control group , and the others were fed with high-fat diet.When the average weight of the high-fat diet rats was 20% as many as the average weight of the control group , we believed the model succeeded .Then the female SD rats were mated and the pregnant rats were randomly divided into 5 groups , 8 rats in each .They were fed with high fat diet during pregnancy and lactation , and the obese pregnant rats were respectively irrigated with 0 mg/kg body weight ( high-fat control ) , 50 mg/kg body weight,100 mg/kg body weight , and 200 mg/kg body weight quercetin .The blank control group was fed with basic diet throughout the experimental period .The birth weight , the weights during the development of d7, d21, d56, and d98, and the expressions of IGF-1 mRNA in livers tissue of the F1 generation were measured.Results:The average birth weight of the F1 generation of high fat diet (HFD) group was sig-nificantly higher than that of the blank control group .Decreased offspring birth weight was observed with the intervention of quercetin , which effect remained to the delectation .The quercetin also reduced the ex-pression of IGF-1 mRNA in livers of the F1 generation , and this effect remained to the adulthood .The 200 mg/kg body weight quercetin was most significantly effective .Conclusion:The averaged birth weight of the F1 generation of obese pregnant rats was significantly increased , and quercetin could effectively in-hibit the expression of IGF-1 mRNA in livers of F1 generation to decrease cell proliferation and cell differentiation.
