目的:观察模拟失重条件下人体尿液代谢组学特征的变化。方法:采用-6°头低位卧床的方法建立人体模拟失重模型,被试者为15例健康成年男性,试验期间统一膳食,分别在卧床固定时间点采集尿液进行拉曼光谱检测,将其所得光谱数据通过PPF-PCA分析,观察卧床条件下各组尿液代谢产物谱的差异以及随时间的变化。结果:按卧床时间区分,被试者尿液代谢产物谱可划分为4种状态,状态1:R-10、R-2、R2;状态2:R4、R7、R14;状态3:R21、R30;状态4:R37、R44、R+7。状态1到状态4之间的差异有逐渐变大的趋势,状态之间的变化点依次为R4、R21、R37。结论:本试验中,被试者尿液代谢产物谱分别在卧床试验的第4天、第21天、第37天发生明显变化,此结果可为后续潜在代谢标志物的筛选以及营养干预措施的制定提供线索。

Objective:To investigate the change of urine metabolic profile in weightlessness-simulated human.Method:Weightlessness was simulated by-6° head-down-tilt bed rest,the normal adult males were selected and the food and work-rest were unified in the experiment. The urine were collected and detected by raman spectra in the fixed time of bed-rest. The spectrum data were analyzed by projection of basing on principal component and fisher criterion method. Result:According to the time of bed-rest,the urine metabolic profile were devided into 4 states which was including the 1st state:R-10,R-2,R2;the 2nd state:R4,R7,R14;the 3rd state:R21,R30 and the 4th state:R37,R44,R+7. From 1st state to 4th state,the difference was gradually decreased and the change points were R4,R21,R37. Conclusion:In the experiment,the urine metabolic profiles were significantly changed in the 4th day,the 21st day,the 37th day,the clew was provided for the selecttion of potential metabolism markers and formulation of nutrition ntervention study in the experimental results.
