通过对土壤微生物总DNA的提取方法改进,获得改进提取法。利用改进提取法与MO BIO公司PowerSoil DNA Isolation Kit的试剂盒法,对4种不同林分的土壤微生物总DNA进行提取验证。结果表明,2种方法均能有效提取森林土壤总DNA,试剂盒法操作简单,DNA的提取质量高,后续PCR效果好,但其提取的DNA量较少且成本昂贵;改进提取法提取的DNA量是试剂盒法的2倍,较经济,但耗时较长,操作繁琐,后续PCR效果与试剂盒法比较相差不大,由于其DNA提取量多,该提取方法较适合应用于土壤宏基因组相关的分子生态学研究中。

Two methods for total deoxyribonucleic acid ( DNA) extraction of forest soil microorganisms were compared-the improved extraction method and PowerSoil DNA Isolation Kit method of MO BIO company , and both methods were used to validate soil microorganisms total DNA extraction for 4 kinds of different forest stands .Results showed that both methods were capable of extracting DNA, effectively.The kit method was easy and the quality of DNA was higher , and better PCR effect , but it was expensive and had less DNA extracted than the improved method .By the improved extraction method 2 times DNA yield was obtained with less cost than by the kit method , but it took long time operating cumbersome , PCR effect of both methods had no significant difference.The improved method can be applied to related research of molecular ecology in soil metagenomic due to the high DNA yield .
