目的 探讨MMP9、MMP14在乳腺癌中的表达及与乳腺癌淋巴管生成、淋巴结转移的关系.方法 应用手工制作组织芯片和免疫组织化学技术(SP法)检测乳腺良性病变、乳腺癌组织中MMP9、MMP14的表达水平.通过免疫组织化学技术(SP法)用D2-40标记淋巴管,进行淋巴管计数.结果 MMP9、MMP14在乳腺良性病变组和乳腺癌组之间比较均有统计学差异(P<0.01).MMP9、MMP14表达与肿瘤大小、导管癌分级、淋巴结转移之间的差异有显著性(P<0.01).乳腺癌周区淋巴管密度(LVD)均值明显高于中心区和乳腺良性病变组的LVD均值,其结果有统计学意义(P<0.01).MMP9、MMP14表达状况与癌周区LVD之间的差异有显著性(P<0.01).结论 MMP9、MMP14与淋巴管生成及淋巴结转移关系密切并成正相关关系.

Objective To explore the association of the expressions of MMP-9 and MMP-14 in breast cancer with lymph node metastasis and lymphangiogenesis. Methods The expression levels of MMP-9 and MMP-14 were detected by hand-made tissue microarray and immunohistochemistry in benign breast diseases and breast cancer. The number of lymphatic vessels marked with D2-40 mark was counted. Results The expressions of MMP-9 and MMP-14 differed significantly between benign breast disease group and breast cancer group ( P< 0.01 ). There were significant difference in the expressions of MMP-9 and MMP-14 based on tumor size, stages of ductal carcinoma, and lymph node metastasis ( P< 0.01 ). The lymphatic vessel density ( LVD )was significantly higher in the paracancerous areas than in the central areasand the benign breast tissues ( P<0.01 ). There were significant differences between the expressions of MMP-9 and MMP-14 and LVD in the paracancerous areas (P < 0.01 ). Conclusions MMP-9 and MMP-14 are positively related with lymphangiogenesis and lymph node metastasis.
