目的：细胞核仁形成区（NORs）由核糖体RNA基因组成，位于此区调控rRNA转录的酸性非组蛋白可被硝酸银染色（Ag-NOR），肿瘤细胞中 Ag-NOR 数目、分布和强度可作为肿瘤诊断、细胞分型和分化分级的重要指标。由于肿瘤细胞相关分子等的毒性作用，患者淋巴细胞NORs区的酸性非组蛋白得以表达。有别于非恶性疾病，并以此进行肿瘤诊断和病情监测。方法北京健尔康医疗设备有限公司提供的KL型肿瘤免疫图像分析系统，5%硝酸银及相应的银染试剂，RPMI1640培养液，普通细胞银染技术对患者外周血淋巴细胞染色。选择35例恶性肿瘤患者及35例健康对照者，经培养、制片、银染、图像分析四个步骤，计算T淋巴细胞核银染面积与细胞面积比值（即 I.S%值）。结果对70例正常者和各种肿瘤患者的淋巴细胞酸性非组蛋白表达活性进行了对比研究，发现恶性疾病与正常者之间有明显差异。35例肿瘤患者中有28例阳性结果，有统计学意义，说明I.S%阳性结果与肿瘤有较密切联系，但并无特异性，这与有关文献报道相符。结论外周血T淋巴细胞Ag-NORs检测，可作为恶性肿瘤诊断及病情监测的重要参考指标。

Objective The nucleolus organizer regions(NORs)are composed by ribosome(RI)RNA genes.Theseregions control and regulate the acor-nonhistone protein transcriptived by rRNA,and also can be stained by argent nitrate (AgNO3)-Ag-NOR.The numbers,distribution and intensity of Ag-NOR in tumor cels(TC)can be used as significant index in the diagnose,celltyping,celldiferentiation ranking of tumor.In case of the toxic efects of related molecule in TC,there are some diferences in the expression of acor-nonhistone protein in the nucleolus organizer regions(NORs)between the patients with malignant tumor and non-malignant tumor,this can be used as a method of the diagnose of tumor and the monitoring of pathogenetic condition.Methods The KL-model image analytical system of tumor immunity was provide by Beijing JIANERKANG medical treatment facility company,Ltd.Other materials included 5%AgNO3 and the proportional silver staining reagent,and the RPMl 640 culture solution.The ordinary silver staining technique was used to stain peripheral-blood lymphocytes of the patients.Results The contrast study of the activity of leukomonocyte acor-nonhistone protein expression were done in 70 health adult and patients with diferent tumor diseases,and remarkable difference was found between the patients with tumor diseases and health people.28 cases were positive results among 35 cases of tumor patients,which with statistical significance. This indicated the I.S% positive result was comparatively closed relation with tumors.This result is in accordance with some related literature reports.Conclusion The detection of Ag-NORs of the peripheral-blood T-lymphocytes can be used as an important reference index in the diagnose of malignant tumor and the monitoring of pathogenetic condition.
