背景 Nocth信号系统在干细胞分化的调控中起着重要作用,但尚没有脐血来源干细胞分化为心肌细胞的机制中Notch作用的报道。目的观察Notch信号在脐血间充质干细胞向心肌细胞分化过程的调控作用。方法人脐血来源干细胞与心肌细胞共培养,实验1组加入Notch激活剂Jagged1,实验2组加入Notch激活剂Jagged1和其抑制剂DAPT,对照组加入PBS液。用免疫组化方法检测干细胞分化为心肌细胞及Notch信号的表达。结果与结论脐血间充质干细胞在体外可分化为心肌细胞,与对照组和实验2组相比,实验1组的干细胞分化为心肌细胞的比率更高,并且Notch1和Jagged1表达增强。说明Notch信号系统在脐血干细胞分化为心肌细胞中起正向调控作用。

BACKGROUND:The Nocth signal system plays an important role in the regulation of the differentiation of stem cel s, but there is no reports of Notch signaling on umbilical cord blood -derived stem cel s differentiated into cardiomyocytes.OBJECTIVE: To Observe Notch signaling regulation of differentiation process in umbilical cord blood mesenchymal stem cel s to myocardial cel .METHODS: Human umbilical cord blood -derived stem cel s were cocultured with cardiac myocytes in viltro.The experimental groups 1 were added Notch activator of Jagged1, The experimental groups 2 were added Jagged1 Notch activator and its inhibitor DAPT, The control group were joined in PBS . The expression of myocardial cel s and Notch signaling in stem cel was detected byimmunohistochemicalmethods .RESULTS AND CONCLUSION: Umbilical cord blood mesenchymal stem cel s were differentiated into myocardial cel s in vitro , compared with the control group and the experimental group 2 , the experimental group 1 were into the higher ratio of myocardial cel s , and Notch1 and Jagged1 expression was enhanced . Notch signaling system in the differentiation of umbilical cord blood stem cel s t into myocardial cel s plays a positive role.
