　　目的：探讨精液体外处理方法及技巧。方法：2010年取卵周期数为1402例实施部分上游法及部分密度梯度离心法；2011年取卵周期数为1667例实施大部分梯度密度离心法+上游法及少部分上游法；2012年取卵周期数为1321例实施全部梯度密度离心+上游法。观察并记录各年份患者受精情况，进行统计学分析，得出结论。结果：本院自2010年-2012年，临床妊娠成功率呈现出逐年上升趋势，结果比较差异具有统计学意义（P<0.05）。结论：应用密度梯度离心+上游序贯法可使精子的运动参数及形态学均得到明显改善，增加患者妊娠率及保障精子核DNA链完整性，是较好的体外处理精液方法，值得临床推广应用。

Objective:To explore the methods and skills of sperm in vitro.Method:1402 cases of patients with semen taken part upstream method and density gradient centrifugation in 2010;1667 cases of patients with semen taken most upstream density gradient centrifugation method and a few of the upstream method in 2011;1321 cases of patients with semen implementing all gradient density centrifugation+upstream method in 2012.The condition of fertilization of patients in different years were observed and recorded,and statistical analysis and concluded.Result:In our hospital from 2010 to 2012, the success rate in patients with pregnancy showed a rising trend year by year,and the difference was statistically significant(P<0.05).Conclusion:The application of density gradient centrifugation+upstream sequential method in clinical practice,not only can improve sperm motion parameters,but also can increase the success rate in patients with pregnancy,the clinical effect is good,it is worth popularizing in clinic.
