[目的]研究缺氧对阴茎海绵体平滑肌细胞(cavernosum smooth muscle cel s ,CCSM)表型转化的影响。[方法]用酶消化法获取SD大鼠的原代CCSM,并用细胞免疫荧光法进行鉴定。将原代CCSM置于缺氧小室中(37℃,5%CO2,1%O2,94%N2)培养,按缺氧时间分为0h组、24h组、48h组、72h组,用western-blot法检测四组的α-SMA、OPN蛋白表达量。[结果]低氧可以诱导CCSMα-SMA蛋白表达下降,且在72h内随着缺氧时间延长差异显著性增加。低氧24h组细胞OPN蛋白表达增加(P<0.05);低氧48h组细胞OPN蛋白表达显著增加(P<0.01);48h后随着时间延长差异可能会减小(P>0.05)。[结论]缺氧可以诱导CCSM由收缩型向合成型转化。

[목적]연구결양대음경해면체평활기세포(cavernosum smooth muscle cel s ,CCSM)표형전화적영향。[방법]용매소화법획취SD대서적원대CCSM,병용세포면역형광법진행감정。장원대CCSM치우결양소실중(37℃,5%CO2,1%O2,94%N2)배양,안결양시간분위0h조、24h조、48h조、72h조,용western-blot법검측사조적α-SMA、OPN단백표체량。[결과]저양가이유도CCSMα-SMA단백표체하강,차재72h내수착결양시간연장차이현저성증가。저양24h조세포OPN단백표체증가(P<0.05);저양48h조세포OPN단백표체현저증가(P<0.01);48h후수착시간연장차이가능회감소(P>0.05)。[결론]결양가이유도CCSM유수축형향합성형전화。
[Objective] To observe the influence of hypoxia on phenotypic modulation in corpus cavernosum smooth muscle cel s(CCSM). [Methods] Pri-mary CCSMC were obtained through enzymatic digestion in SD rat cavernous tissue, identified by immunofluorescence method. Primary CCSM were cultured in hypoxia(37℃, 5%CO2, 1%O2, 94%N2) with different time(0h, 24h, 48h, 72h). Western-blot technique was used to detect the protein expres-sion of α-SMA, OPN in that four groups of hypoxia-induced time. [Results] Hypoxia could down-regulate the protein expression of α-SMA, which was increased markedly with longer anoxia in 72h;the hypoxia in 24h increased the cel OPN protein expression; as wel as in the group of 48h;the dif-ference may decrease after 48h with more time.[Conclusion] Hypoxia can induce the transformation of CCSM from contractile to synthetic.
