[目的]探讨苗药皮部熏洗法治疗骨关节炎(Osteoarthritis,OA)的部分机理。[方法]取42只健康清洁级大鼠,随机选取8只不造模作为正常对照组,其余34只采用4%木瓜蛋白酶关节腔注射建立大鼠OA模型,一周后随机选取2只检测模型成功与否,将造模成功的32只大鼠随机分为模型组、熏组、洗组和熏洗组,治疗4周后处死动物,切取关节,采用HE染色进行关节软骨病理评分(Mankin's评分),免疫组化染色法测定PCNA及TGF-β1的表达。[结果]Mankin's评分结果显示:与模型组比较,各治疗组均降低Mankin's评分(P<0.05,P<0.01),且熏洗组与熏组、洗组相比差异显著(P<0.01)。正常组有少量PCNA和TGF-β1阳性细胞的表达,模型组中表达明显,治疗组均比模型组升高(P<0.05),其中熏洗组与模型组比差异显著(P<0.01)。[结论](1)通过苗药皮部熏洗的干预,对OA模型大鼠的痛行为及膝关节活动有明显的改善作用。(2)苗药皮部熏洗通过上调软骨PCNA和TGF-β1的表达,促进软骨细胞的增殖,加速软骨的修复,达到防治OA的目的。(3)熏洗法治疗大鼠OA比单独运用熏法、洗法效果更明显。

[목적]탐토묘약피부훈세법치료골관절염(Osteoarthritis,OA)적부분궤리。[방법]취42지건강청길급대서,수궤선취8지불조모작위정상대조조,기여34지채용4%목과단백매관절강주사건립대서OA모형,일주후수궤선취2지검측모형성공여부,장조모성공적32지대서수궤분위모형조、훈조、세조화훈세조,치료4주후처사동물,절취관절,채용HE염색진행관절연골병리평분(Mankin's평분),면역조화염색법측정PCNA급TGF-β1적표체。[결과]Mankin's평분결과현시:여모형조비교,각치료조균강저Mankin's평분(P<0.05,P<0.01),차훈세조여훈조、세조상비차이현저(P<0.01)。정상조유소량PCNA화TGF-β1양성세포적표체,모형조중표체명현,치료조균비모형조승고(P<0.05),기중훈세조여모형조비차이현저(P<0.01)。[결론](1)통과묘약피부훈세적간예,대OA모형대서적통행위급슬관절활동유명현적개선작용。(2)묘약피부훈세통과상조연골PCNA화TGF-β1적표체,촉진연골세포적증식,가속연골적수복,체도방치OA적목적。(3)훈세법치료대서OA비단독운용훈법、세법효과경명현。
[Objective] Investigate Miao medicine fumigation and washing treating skin osteoarthriti(Osteoarthritis, OA) for the partial mechanism.[Methods] As for the 42 healthy rats, the randomly selected 8 of them were not built into a model group but as a normal control group, and the remaining 34 were adopted to establish the rat model, regarded as OA model of 4% Papuan inter-articulation injection. After checking whether the randomly selected 2 de-tection models were successful or not a week later, we adopted 32 model rats and randomly divided them in the model group, smoking group, washing and fumigation group and then after 4 weeks of treatment, we kil ed them and cut off their joints, using the HE staining for Mankin's testing, the Im-munohistochemistry staining method of PCNA and TGF-β1 in the determination of staining method. [Results] The Mankin's testing results showd:com-pared with the model group, the fumigation and washing group had significant difference. The normal group expressed a smal amount of PCNA and the expression of TGF-β1 positive cel s in the model group, the treatment group were higher than the model group( P<0.05), wherein the fumigation group compared with model group had significant difference (P<0.01).[Conclusion] (1) The intervention by Miao medicine fumigation on skin, knee Pain activi-ty behavior of OA model rats had obvious improvement.(2) Miao medicine fumigation regulating cartilage skin PCNA and TGF-β1, promoted the prolif-eration of chondrocyte, accelerated the cartilage repair, and achievd the purpose of preventing OA. (3) The treatment of fumigation and washing group were more obvious than the single fumigation group and washing group.
