目的:建立高效液相色谱法测定大鼠心肌组织中腺苷的含量。方法 SD大鼠随机分为心肌缺血再灌注组( IRI)、106 Treg细胞转输+IRI组、107Treg细胞转输+IRI 组和假手术对照组。色谱柱为 VP-ODS C18柱(250mm ×4.6mm,5μm);流动相为甲醇:10mmol· L-1乙酸铵(10∶90);流速为1.0mL· min -1;柱温25℃;检测波长260nm;进样量20μL。结果腺苷的标准液与峰面积均呈良好的线性关系(r=1.000),平均回收率分别为100.1%。日内和日间精密度分别在4.56%和4.84%以下。结论本法操作简便、准确、快速,可以用于测定大鼠心肌组织中腺苷的含量。

목적:건립고효액상색보법측정대서심기조직중선감적함량。방법 SD대서수궤분위심기결혈재관주조( IRI)、106 Treg세포전수+IRI조、107Treg세포전수+IRI 조화가수술대조조。색보주위 VP-ODS C18주(250mm ×4.6mm,5μm);류동상위갑순:10mmol· L-1을산안(10∶90);류속위1.0mL· min -1;주온25℃;검측파장260nm;진양량20μL。결과선감적표준액여봉면적균정량호적선성관계(r=1.000),평균회수솔분별위100.1%。일내화일간정밀도분별재4.56%화4.84%이하。결론본법조작간편、준학、쾌속,가이용우측정대서심기조직중선감적함량。
OBJECTIVE To develop a high performance liquid chromatography ( HPLC) method to determine the content of adenosine in myocardium of rats.METHODS Male SD rats were divided into ischemia-reperfusion group,106 Treg cell transmission treatmen +IRI group、107 Treg cell transmission treatment +IRI group and shamoper-ated group.HPLC analysis was preformed using a VP-ODS C18 column (250mm ×4.6mm,5μm).The mobile phase consisted of methanol and 10 mmol· L-1 ammonium acetate with ratio of 10 to 90.The flow rate was 1mL· min-1 and the temperature of column was about 25℃.The detection wavelength was 260 nm and the injection volume was 20μL.RESULTS The calibration curve showed good linearity ( r=1.000 ) and the average recovery rates of aden-osine was 100.1%.The within-day and between-day precisions were less than 4.56% and 4.84%, respective-ly.CONCLUSION This analytical method is simple , rapid and specific in detecting the content of adenosine in myocardium of rats with myocardial ischemia.
