采用电子吸收光谱、粘度测定和电化学等方法研究了氨基黑10B(AB)与鲱鱼精DNA在B-R缓冲溶液中的相互作用。电子吸收光谱实验表明,DNA的加入能引起AB在618 nm处的最大吸收峰出现增色效应,说明二者的结合不属于典型的静电或嵌插结合模式,而可能通过沟槽方式结合,且伴随有氢键作用。粘度法实验证实了这一结果。电化学实验进一步表明DNA的加入能引起AB分子的扩散速率降低,从而导致AB的氧化峰电流出现减小。同时考察了AB与DNA结合前后的电子转移速率,结果表明AB与DNA之间形成了一种非电活性的超分子复合物。

The interaction between amide black 10B (AB) and DNA in B-R buffer solution was investigated by the electronic absorption spectra, viscometry and electrochemistry. The characteristic absorption peak at 618 nm of AB showed obvious hyperchromic effect after interaction with DNA, suggesting that the interaction of AB with DNA was neither ascribed to intercalation nor eleotrostatic action, but via a groove binding mode with some hydrogen bonding, which was testified by viscosity measurement. Electrochemical experiments further suggested that the diffusion rate of AB was lowed after interaction with DNA, resulted in the decrease of oxidation peak currents. The results of electron-transferred rate suggested that an electro-inactive complex was formed between AB and DNA.
