利用13条ISSR引物分析了70份四川地方性晾晒烟材料的遗传多样性.共扩增出107条带,平均每个引物扩增8.23条,引物的平均多态性比率为90.37%,聚类分析表明,品种间遗传相似指数的范围在0.46—0.87,平均遗传相似性系数为0.646.这表明其遗传差异不大.其中遗传相似性最大的品种为达州的土烟与新都的黑牛皮,遗传相似指数为0.46;遗传相似性最小的品种为什邡的红毛柳和巴毛柳,遗传相似指数为0.87.聚类和主成分分析一致表明,70份材料在遗传相似系数为0.646处分为5类,并且四川晒烟种质资源遗传多样性与其地理来源相关.ISSR标记能高效揭示亲缘关系很近的晾晒烟种质资源的亲缘关系,因此适用于烟草品种鉴定和遗传多样性研究.

Genetic diversity of 70 Sichuan sun-cured tobacco cuhivars was analyzed by inter simple sequence repeats (ISSR). 107 bands were identified using agarose gel eleetrophoresis. The average number of amplified DNA bands was 8.23 per primer, and the rate of polymorphic bands was 90.37%. Genetic similarity ranged from 0.46 to 0.87 through cluster analysis with UPGMA, which suggested the genetic diversity among these germplasms not obvious, and the average genetic similarity index was 0.646. Genetic similarity index between Hong maoliu and Ba maoliu was 0.87, indicating that their genetic diversity was similar, but genetic similarity index between Tu yan from Da zhou and Hei niupi from Xin du was 0.46, showing more difference of their genetic diversity. Cluster analysis and principal component analysis (PCA) were carried out. The results showed that the 70 Sichuan sun-cured tobacco cultivars were divided into five groups, deeply associated with their geographical distribution. ISSR analysis was suitable for variety identification and the research on genetic diversity of tobacco germplasm.
