本试验采用冷热两种消化法对羊睾丸组织进行消化分离,分别进行原代细胞的培养,细胞长成单层后进行接毒,分别收获毒液配苗,并对半成品、成品进行测毒,结果表明,冷热两种消化方法相比较,冷消化法的作用较温和,对原代细胞的损伤较小,细胞活力较高,可明显的提高生产效率并大大降低生产成本。因此,这种方法可以大力推广,值得同行借鉴。

In this study, we chose warm and cold digest methods to separate hircine testis tissue, and then cultured primary cell respectively. Cells were infected by goat pox virus to harvest venom and prepare vaccine when them growed into a monolayer. Compared warm and cold enzyme digestion methods, results showed that cold digestive reaction was more milder, damaged less to primary cells and had higher cell viability through tests of semi-finished vaccine's poison determination and finished vaccine's virus attack. It is evident that the product significantly can be improved and the production costs can be reduced using the same weight of testis tissue to cultivate cells by adding different amounts of nutrient. Therefore, above methods can be promoted and worthy of peer learning.
