目的:探讨浒苔多糖脱蛋白脱盐的方法。方法:利用蛋白酶水解浒苔多糖的蛋白质,通过正交试验优选蛋白酶水解工艺条件;利用透析袋流水透析脱盐,通过正交试验优选透析工艺条件。结果:中性蛋白酶脱蛋白效果优于Sevag法和三氯乙酸法,中性蛋白酶脱蛋白较佳工艺条件为:加酶量0.12g/10mL、pH6.0、反应温度45℃、反应时间1h,在此条件下,蛋白质脱除率达52.36%。透析脱盐的较佳条件为:7000Da、透析时间8h、浒苔多糖溶液浓度6%,在此条件下,灰分脱除率为76.95%。(除杂后,浒苔多糖蛋白质含量为3.43%,灰分含量为8.04%,多糖损失率为18.96%)。结论:蛋白酶法脱蛋白质技术和透析袋流水透析脱盐技术,对浒苔多糖的脱蛋白脱盐效果较好,有一定的推广潜力。

목적:탐토호태다당탈단백탈염적방법。방법:이용단백매수해호태다당적단백질,통과정교시험우선단백매수해공예조건;이용투석대류수투석탈염,통과정교시험우선투석공예조건。결과:중성단백매탈단백효과우우Sevag법화삼록을산법,중성단백매탈단백교가공예조건위:가매량0.12g/10mL、pH6.0、반응온도45℃、반응시간1h,재차조건하,단백질탈제솔체52.36%。투석탈염적교가조건위:7000Da、투석시간8h、호태다당용액농도6%,재차조건하,회분탈제솔위76.95%。(제잡후,호태다당단백질함량위3.43%,회분함량위8.04%,다당손실솔위18.96%)。결론:단백매법탈단백질기술화투석대류수투석탈염기술,대호태다당적탈단백탈염효과교호,유일정적추엄잠력。
【Objective】 To study the method of de-protein and desalting process for enteromorpha polysaccharides.【Method】The best method of enzymatic de-protein process was screened from amino-acid nitrogen.The proper method of run water dialysis was chosen from dry ash content and polysaccharides loss rate.【Result】De-protein effect of neutral protease was better than that of papain method,Sevag method and trichloroacetic acid method.The best condition of neutral protease de-protein process was adding 0.12g/10mL of lipase amounts into the extracting liquid,adjusting the pH to 6.0,then keeping it in the constant temperature bath 45℃ for 1h.The proper method of run water dialysis was keeping 6% enteromorpha extracting liquid in the 7?000Da dialysis bag for 8h.The protein content of purer enteromorpha polysaccharides was 3.43%,ash content was 8.04%,and polysaccharides loss rate was 18.96%.【Conclusion】 Neutral protease method and of run water dialysis method are the better ways for de-protein effect and desalting process,which can be used universally.
