目的:构建低氧诱导的非酒精性脂肪性肝炎动物模型。方法50只大鼠分为正常组、低氧组、高脂组、低氧高脂组4组,分别给予正常饮食、腹腔内注射亚硝酸钠、高脂饮食、腹腔内注射亚硝酸钠+高脂饮食。检测4组大鼠生化指标(ALT、AST、AKP、TG)、透明质酸(HA)、动脉高铁血红蛋白及氧分压,并行肝组织HE与Masson染色及NASH评分。结果低氧高脂组大鼠的ALT、AST、AKP、TG、HA明显升高,NASH评分明显增加,肝组织可见明显的脂肪变性、炎症细胞浸润及纤维化。结论高脂饮食大鼠诱导单纯脂肪肝基础上给予腹腔注射亚硝酸钠能够建立低氧诱导的非酒精性脂肪性肝炎动物模型。

목적:구건저양유도적비주정성지방성간염동물모형。방법50지대서분위정상조、저양조、고지조、저양고지조4조,분별급여정상음식、복강내주사아초산납、고지음식、복강내주사아초산납+고지음식。검측4조대서생화지표(ALT、AST、AKP、TG)、투명질산(HA)、동맥고철혈홍단백급양분압,병행간조직HE여Masson염색급NASH평분。결과저양고지조대서적ALT、AST、AKP、TG、HA명현승고,NASH평분명현증가,간조직가견명현적지방변성、염증세포침윤급섬유화。결론고지음식대서유도단순지방간기출상급여복강주사아초산납능구건립저양유도적비주정성지방성간염동물모형。
Objective To establish a animal model for hypoxia- induced non- alcoholic steatohepatitis. Methods Rats were randomly assigned to a normal diet (control group), normal diet+intraperitoneal injection of sodium nitrite (hypoxia group), high fat diet (high fat group), intraperitoneal injection of sodium nitrite+high fat diet (hypoxia+high fat group), respectively. Bio-chemical markers including alanine aminotransferase (ALT), aspartate aminotransferase (AST), total triglycerides (TG) and liver fi-brosis marker hyaluronic acid (HA), methemoglobin and arterial oxygen partial pressure were detected. In addition, liver tissue HE and Masson staining and NASH score were performed. Results The levels of ALT, AST, TG, HA and NASH score were signifi-cantly increased in hypoxia+high fat group. Steatosis, inflammatory infiltration and fibrosis was observed in liver tissues of hy-poxia+ high fat group. Conclusion Intraperitoneal injection of sodium nitrite can establish a hypoxia- induced non- alcoholic steatohepatitis animal model on the basis of high fat diet.
