目的:探讨Tol 样受体(TLRs)2、3、4、9在不同孕期胎盘组织中的表达及其与自发性早产的关系。方法选择自发性早产患者的胎盘样本38份作为早产组(其中孕28-34周和孕35~36周各19份)。选择同期正常妊娠自然分娩产妇的胎盘样本38份(孕37-40周)作为对照组。采用real- time PCR法检测两组胎盘中TLR 2、3、4、9 mRNA的表达,并用Western blot法检测胎盘中TLRs 2、3、4、9蛋白的表达,对结果进行统计学分析。结果(1)两组孕产妇胎盘组织中均可检测到TLRs 2、3、4、9的表达。(2)早产组胎盘组织TLR 2、3、9 mRNA的表达均高于对照组,两者间差异有统计学意义(均P<0.05);TLR4 mRNA的表达两组间无统计学差异(P>0.05)。(3)早产组TLR 2、3、4、9的蛋白表达均高于对照组,两者差异均有统计学意义(均P<0.05)。(4)早产组中,28~34孕周组和35~36孕周组胎盘中TLRs mRNA表达量均无统计学差异(均P>0.05)。结论早产患者胎盘组织中TLRs过度表达,提示TLRs介导的母胎界面的免疫反应可能是启动自发性早产的重要因素。

목적:탐토Tol 양수체(TLRs)2、3、4、9재불동잉기태반조직중적표체급기여자발성조산적관계。방법선택자발성조산환자적태반양본38빈작위조산조(기중잉28-34주화잉35~36주각19빈)。선택동기정상임신자연분면산부적태반양본38빈(잉37-40주)작위대조조。채용real- time PCR법검측량조태반중TLR 2、3、4、9 mRNA적표체,병용Western blot법검측태반중TLRs 2、3、4、9단백적표체,대결과진행통계학분석。결과(1)량조잉산부태반조직중균가검측도TLRs 2、3、4、9적표체。(2)조산조태반조직TLR 2、3、9 mRNA적표체균고우대조조,량자간차이유통계학의의(균P<0.05);TLR4 mRNA적표체량조간무통계학차이(P>0.05)。(3)조산조TLR 2、3、4、9적단백표체균고우대조조,량자차이균유통계학의의(균P<0.05)。(4)조산조중,28~34잉주조화35~36잉주조태반중TLRs mRNA표체량균무통계학차이(균P>0.05)。결론조산환자태반조직중TLRs과도표체,제시TLRs개도적모태계면적면역반응가능시계동자발성조산적중요인소。
Objective To investigate the expression of Tol- like receptors (TLRs) in placenta and its relationship with spontaneous preterm labor. Methods Thirty eight placental samples were col ected from patients with spontaneous preterm labor (preterm group), including 19 of 28~34 week gestation, and 19 of 35~36 week gestation. Thirty eight placental samples from normal pregnancy and natural delivery (37~40 week) served as control group. The mRNA expressions of TLR 2, 3, 4, and 9 were detected by real- time quantitative polymerase chain reaction (RT- PCR);and the protein expression of TLR2, 3, 4, and 9 were evaluated by Western blot. Results The mRNA expressions of TLR2, 3, 9 in preterm group were significantly higher than those in control group(P<0.05), but not for TLR4 (P>0.05). The protein expressions of TLR2, 3, 4 and 9 also showed higher in preterm group than those in control group (P<0.05). Conclusion The over- expressed TLRs in preterm placental tissue sug-gests that the immune response mediated by TLRs in the maternal- fetal interface may be important factors to initiate sponta-neous preterm labor.
