为了建立盐胁迫条件下小麦叶片蛋白的双向电泳研究方法,进一步获取小麦叶片响应盐胁迫的差异蛋白图谱,以河南省主导小麦品种‘周麦18’为实验材料,利用NaCl溶液模拟盐分胁迫,在建立小麦幼苗双向电泳技术和图像分析体系的基础上,对盐胁迫下的小麦幼苗进行差异蛋白组学初步分析。结果表明:72 h 1.0%NaCl溶液胁迫下,小麦幼苗蛋白表达组发生了变化;其中共有332个点得到匹配,获得61个差异表达的蛋白点,29个表达量显著上调,32个显著下调。这表明,在盐胁迫环境条件下,植物为了满足对NaCl的需求,可能开启和关闭了体内的一些相关代谢途径。
為瞭建立鹽脅迫條件下小麥葉片蛋白的雙嚮電泳研究方法,進一步穫取小麥葉片響應鹽脅迫的差異蛋白圖譜,以河南省主導小麥品種‘週麥18’為實驗材料,利用NaCl溶液模擬鹽分脅迫,在建立小麥幼苗雙嚮電泳技術和圖像分析體繫的基礎上,對鹽脅迫下的小麥幼苗進行差異蛋白組學初步分析。結果錶明:72 h 1.0%NaCl溶液脅迫下,小麥幼苗蛋白錶達組髮生瞭變化;其中共有332箇點得到匹配,穫得61箇差異錶達的蛋白點,29箇錶達量顯著上調,32箇顯著下調。這錶明,在鹽脅迫環境條件下,植物為瞭滿足對NaCl的需求,可能開啟和關閉瞭體內的一些相關代謝途徑。

The aims were to set up an approach on two-dimensional electrophoresis for wheat leaf proteins under salt stress, and obtain the discrepancy-expressed protein map responded to high-salt content in soil. For this study, the experimental material was a main wheat cultivar‘Zhoumai-18’in Henan, NaCl solution was utilized to imitate the salt stress. Based on the establishment of two dimensional electrophoresis and image analysis, the proteomic difference of wheat seedlings under salt stress was initially analyzed. The results showed that, the whole proteins extracted from young seedlings cultured in pots under 1.0%NaCl for 72 hours were changed. 332 proteins were matched and 61 proteins showed differently expressed under salt stress, with 29 up and 32 down. This indicated plants maybe turn some relevant metabolic pathways inside them on and off under salt stress in order to satisfy its need for NaCl.
