目的:探讨在大鼠睾丸扭转/复位过程中CD4+T细胞亚群Th1/Th2失衡对生殖细胞凋亡的影响。方法 SD雄性成年大鼠30只,随机分成假手术组、睾丸扭转组和复位组,每组10只。通过建立左侧睾丸扭转和扭转复位动物模型,在手术后24h分别取材,血清酶联免疫法(ELISA)检测血清干扰素γ(IFN-γ)、白细胞介素12(IL-12)、白细胞介素4(IL-4)和白细胞介素10(IL-10)水平;原位缺口末端标记法(TUNEL法)检测各组左侧睾丸生殖细胞凋亡指数。结果与假手术组相比,扭转组和扭转复位组与Th1细胞相关的细胞因子IFN-γ、IL-12水平升高明显(P<0.05),Th2细胞相关的细胞因子IL-4和IL-10水平下降(P<0.05);生殖细胞凋亡指数增加。结论在大鼠睾丸扭转和复位过程中CD4+T细胞亚群偏移与生殖细胞凋亡存在相关性。

목적:탐토재대서고환뉴전/복위과정중CD4+T세포아군Th1/Th2실형대생식세포조망적영향。방법 SD웅성성년대서30지,수궤분성가수술조、고환뉴전조화복위조,매조10지。통과건립좌측고환뉴전화뉴전복위동물모형,재수술후24h분별취재,혈청매련면역법(ELISA)검측혈청간우소γ(IFN-γ)、백세포개소12(IL-12)、백세포개소4(IL-4)화백세포개소10(IL-10)수평;원위결구말단표기법(TUNEL법)검측각조좌측고환생식세포조망지수。결과여가수술조상비,뉴전조화뉴전복위조여Th1세포상관적세포인자IFN-γ、IL-12수평승고명현(P<0.05),Th2세포상관적세포인자IL-4화IL-10수평하강(P<0.05);생식세포조망지수증가。결론재대서고환뉴전화복위과정중CD4+T세포아군편이여생식세포조망존재상관성。
Objective To investigate the effects of the unbalance of CD4+T cell subgroups on germ cell apoptosis during rat testicular torsion. Methods Male SD rats were randomly divided into three groups and there were 10 rats in each group. For rats in sham operation group, they served as the control;For rats in left unilateral testicular torsion group, their left testes were rotated 720 degree for 2 hours;For rats in left testicular torsion reset group, their testes were reseted after 2 hours testicular torsion. At 24 hours after operation, levels of serum cytokines IFN-γ, IL-12, IL-4 and IL-10 were measured by ELISA. Germ cell apoptosis index in left testes of the rats was determined by TUNEL. Results Compared with that of the sham operation group, the levels of Th1 type cytokines were significantly increased in torsion and detorsion groups(P<0.05). The levels of Th2 type cytokines such as IL-4, IL-10 were obviously lower in torsion and detorsion groups than that in sham operation group (P<0.05). CD4+T cell subgroups was unbalance during testicular torsion in rats. The apoptosis index of germ cells was significantly higher in torsion and detorsion groups than that of sham operation group. Conclusion CD4+T cell subgroups is unbalance in ischemia reperfusion injury in rat testis, which may be associated with germ cell apoptosis.
