以RP59500为试验菌株,在15 L发酵罐内,研究了普那霉素的发酵工艺。通过单因素条件试验确定了其生物合成的最适温度、接种量。优化了其发酵培养基,并进行了发酵中间补料试验,确定了最佳补料工艺。试验结果表明:以3%淀粉、1.7%蔗糖、1%黄豆粉、1.5%酵母粉、0.5%葡萄糖、0.01%KH2PO4、0.2%MgSO4、0.2%泡敌和0.4%CaCO3的发酵培养基配方,以接种量4.0%,26℃下发酵,在发酵21 h时补加氨基酸,普那霉素的发酵效价达3 000 U/mL。

The fermentation techology of pristinamycin with RP59500 as experimental strains was studied in a 15 L fermentor. Through single-factor experiments, the optimum fermentation temperature, inoculation volume were obtained. The fermentation medium composition was optimized.The titer of pristinamycin increased greatly by using feeding fermentation. The results showed that the titer of pristinamycin reached 3 000 μ/mL with 4.0 % inoculation volume at 26 ℃. The optimal medium contains 3% starch, 1.7% sucrose, 1% soybean powder, 1.5% yeast powder, 0.5% glucose, 0.01% KH2PO4,0.2% MgSO4, 0.2% defoaming agent, 0.4% CaCO3, and the amino acid was fed after it had been fermentated for 21 h.
