目的:观察茯苓多糖体外对小鼠精子膜功能、精子DNA 和存活率的保护作用。方法取小鼠精子制备精子悬液,并分为正常组(PBS),生理盐水组,维生素C组(阳性药对照组)和茯苓多糖低、中、高剂量组(0.5mg/mL,1.0mg/mL和5.0mg/mL)。各对照物及茯苓多糖分别与精子悬液共同孵育后,检测精子膜完整性、DNA损伤程度及存活率,通过精子全部低渗膨胀试验(HOS)评估精子膜功能,精子染色质扩散(SCD)实验检测精子DNA完整性,并与已知的抗氧化剂维生素C对照。结果低、中、高浓度的茯苓多糖在相同的条件下均可增加精子存活时间,提高精子存活率和活力,降低精子DNA损伤比率,对精子膜功能和DNA均具有一定的保护作用。其中1.0mg/mL和5.0mg/mL(中、高)的茯苓多糖组作用明显优于维生素C组(P<0.05或P<0.01)。结论茯苓多糖对小鼠精子膜和DNA有明显的保护作用,并可延长精子的体外存活时间。

목적:관찰복령다당체외대소서정자막공능、정자DNA 화존활솔적보호작용。방법취소서정자제비정자현액,병분위정상조(PBS),생리염수조,유생소C조(양성약대조조)화복령다당저、중、고제량조(0.5mg/mL,1.0mg/mL화5.0mg/mL)。각대조물급복령다당분별여정자현액공동부육후,검측정자막완정성、DNA손상정도급존활솔,통과정자전부저삼팽창시험(HOS)평고정자막공능,정자염색질확산(SCD)실험검측정자DNA완정성,병여이지적항양화제유생소C대조。결과저、중、고농도적복령다당재상동적조건하균가증가정자존활시간,제고정자존활솔화활력,강저정자DNA손상비솔,대정자막공능화DNA균구유일정적보호작용。기중1.0mg/mL화5.0mg/mL(중、고)적복령다당조작용명현우우유생소C조(P<0.05혹P<0.01)。결론복령다당대소서정자막화DNA유명현적보호작용,병가연장정자적체외존활시간。
Objective To explore the protective effects of crude extract from Poria cocos on mouse sperm membrane, sperm DNA and viability in vitro. Methods Mice sperm suspension was prepared in PBS, and divided into normal group (PBS), and normal saline group, positive control group (vitamin C) and the low, medium and high dose group(treated with crude extractings from poria cocosuse using a dose of 0.5mg/mL, 1.0mg/mL and 5.0 mg/mL). After the sperms were incubated with the crude extracts, their DNA damaged level and membrane integrity were detected by the improved Eosin-Y (EY) staining, hypo-osmotic swelling (HOS) test and the sperm chromatin dispersion (SCD) test respectively. Results The low, medium and high dosage of crude extractings all prolonged sperm survival time and improved the sperm vitality, reduced the percentage of sperm DNA damage in vitro. Of which, middle and high dose of crude extractings were significantly better than vitamin C group in sperm vitality and sperm DNA damaged level (P<0.05 or P<0.01). Conclusion The crude extracts from poria cocos had significant protective effects on mice sperm membrane and DNA.
