根据成人疾病发育起源的假说,早期发育中的不利影响会导致永久的生理和代谢改变,导致成年后疾病风险增长,建立了大鼠PCOS动物模型,为PCOS的发病机制及治疗方法的研究提供工具.实验采用方法为:将孕鼠分为雄激素组和正常对照组,每组5只,于孕鼠第10d起,雄激素组每天每只注射二氢睾酮1.2mg,对照组注射同体积的生理盐水,待2组仔鼠(正常组13只,二氢睾酮组16只)出生后生长至14~16周,处死,取卵巢观察2组仔鼠卵巢形态,取静脉血测血性激素变化、空腹血糖、胰岛素水平及血脂变化.研究结果表明:雄激素组仔鼠卵巢囊性改变、高雄激素血症、血糖增高以及性周期不规则,与人的多囊卵巢综合症部分特征相似.

According to the disease development hypothesis,adverse effects in early stage lead to permanent changes in physiology and metabolism,resulting in increased risk of suffering diseases in adulthood.In this report,we build a rat PCOS model to gain a deeper understanding of the pathogenesis of PCOS and to evaluate treatment alternatives.Pregnant rats were randomly divided into two experimental groups(n=13,DHT: n=16),on the 15th day of pregnancy,1.2 mg DHT was injected daily with identical volume physiologic saline.The two Pups were raised to 14-16 weeks followed by life extermination,at which time the ovaries were extracted for an array of observations including ovarian morphology,blood gonadal hormone concentration,glucose concentration.The insulin concentrations were tested,and the lipid metabolism was measured.It is found that the DHT rats show the increase in the number of cystic follicles,hyperandrogenic status,hyperglycemia and irregular cycles.The proposed models are expected to make some contributions to the studies of PCOS.
