为探讨镉对原代大鼠肝细胞的毒性作用。用二步灌流法获得大鼠肝细胞,肝细胞暴露于浓度为2.5、5.0、10.0、20.0μmoL/L的醋酸镉中12、24 h,应用四甲基偶氮噻唑蓝(MTT)比色法检测醋酸镉对肝细胞存活率的影响及培养上清液中乳酸脱氢酶(LDH)的活性变化。结果表明,肝细胞暴露于浓度为2.5、5.0、10.0、20.0μmoL/L的醋酸镉中,细胞相对存活率显著下降(P〈0.01),LDH的释放量增加,且具有剂量效应关系。提示一定浓度的醋酸镉可引起肝细胞的毒性损伤。

위탐토력대원대대서간세포적독성작용。용이보관류법획득대서간세포,간세포폭로우농도위2.5、5.0、10.0、20.0μmoL/L적작산력중12、24 h,응용사갑기우담새서람(MTT)비색법검측작산력대간세포존활솔적영향급배양상청액중유산탈경매(LDH)적활성변화。결과표명,간세포폭로우농도위2.5、5.0、10.0、20.0μmoL/L적작산력중,세포상대존활솔현저하강(P〈0.01),LDH적석방량증가,차구유제량효응관계。제시일정농도적작산력가인기간세포적독성손상。
In order to study the toxical effects of cadmium on primary culture hepatocytes in rats. Rat hepatocytes were isolated by a two step perfusion technique, the hepatocytes were incubated with cadmium acetate (2.5 μmoL/L, 5.0 μmoL/L, 10.0 μmoL/L and 20.0 μmoL/L)for 12 h and 24 h, methyhhiazolyldiphenyl-tetrazolium bromide (MTT)technique was used to determine hepatocytes cell viability in this study. The content of lactate dehydrogenase (LDH)in hepatocyte primary culture fluid were determined. The results showed that cell viability decreased significantly (P〈0.01) in a dose dependent manner after exposure to 2.5 μmoL/L,5.0 μmoL/L, 10.0 μmoL/L and 20.0 μmol_/L cadmium acetate,the activities of LDH increased. It suggested that cadmium acetate may induce the damage of hepatocytes at certain doses.
