探讨红车轴草异黄酮提取物(red clover extract,RCE)对雄性db/db 2型糖尿病小鼠糖脂代谢的影响。选用6周龄db/db实验小鼠,随机分成三组,每组4只。每日一次定时定量灌胃,每周测其血糖水平。结果表明,用10和50 mg/kg/day剂量的RCE处理小鼠5 w后,血糖水平分别比未处理组显著降低了40.90%(P〈0.01)和50.38%(P〈0.001),血清甘油三酯比未处理组降低了47.57%(P〈0.01)和43.47%(P〈0.01),血清总胆固醇比未处理组降低了35.59%(P〈0.05)和35.19%(P〈0.05)。同时实时荧光定量RT-PCR结果显示50 mg/kg/day RCE处理组db/db小鼠肝组织APOC3的mRNA表达水平比未处理组显著下降了54.9%(P〈0.05)。这些结果说明了红车轴草异黄酮提取物可能通过抑制肝APOC3的表达改善2型糖尿病小鼠的血脂水平。

To investigate the effect of red clover extract(RCE) on the blood glucose and lipid metabolism of type 2 diabetic db/db mice,male db/db mice were randomly divided into three groups: db/db mice,db/db + 10 mg /kg/day RCE treated,db/db + 50 mg /kg/day RCE treated.The blood glucose levels of db/db mice receiving 10 mg/kg/day or 50 mg/kg/day RCE for 5 weeks significantly decreased by 40.90%(P0.01) and 50.38%(P0.001) respectively,serum triglycerides decreased by 47.57%(P0.01) and 43.47%(P0.01) respectively,and serum total cholesterols decreased by 35.59%(P0.05) and 35.19%(P0.05) respectively.Moreover,the mRNA expression of hepatic apolipoprotein C3(APOC3) were determined by quantitative real time RT-PCR and 50 mg/kg/day RCE treatment was found to significantly down-regulate APOC3 mRNA expression by 54.9%(P0.05).Our results suggest that RCE significantly improves lipid homeostasis in db/db diabetic mice by down-regulating hepatic APOC3 expression.
