为保护野生资源和满足药用栽培的需求,以隔山消的嫩茎为材料,采用组织培养的方法,进行了愈伤组织诱导和分化、不定芽生根、试管苗的生根继代增殖、移栽和定植的研究,建立起嫩茎无性系。结果证明:MS+ZT 0.2 mg/L+2,4-D 2.0 mg/L是嫩茎愈伤组织生长培养的理想培养基;MS+AgNO30.6 mg/L+NAA 0.1mg/L+ZT 0.6 mg/L是愈伤组织分化培养的理想培养基;1/2MS+ABT2号0.8 mg/L+IAA 0.3 mg/L是不定芽生根培养的理想培养基;1/2MS+ABT2号0.6 mg/L+IAA 0.2 mg/L是试管苗生根继代增殖培养的理想培养基;试管苗移栽成活率为91.9%;定植成活率为98%;定植的试管苗长势旺盛、根系非常发达,保持了隔山消的所有植物学性状,翌年鲜块根增收37%。

In order to preserve the wild resources and meet the needs of medical planting, with the method of tissue culture, tender stems of Cynanchum wilfordii were used as material to do the re- search on callus inducement and differentiation, rooting of adventitious buds, rooting multiplication of tube seedlings, ransplantation of tube seedlings and stable planting and established the clone from tender stems. The results show that the optimum medium for callus induction was MS + ZT 0.2 mg/L + 2,4-D2.0 mg/L, MS + AgNO3 0.6 mg/L + NAA 0.1 mg/L + ZT 0.6 mg/L was suitable for callus differentiation, 1/2MS + NO. ABT2 0.8 mg/L + IAA 0.3 mg/L was the best rooting medium for ad- ventitious buds, the ideal medium for rooting multiplication of tube seedlings was 1/2MS + NO. ABT2 0.6 mg/L + IAA 0.2 mg/L,the transplanting survival rate of tube seedlings was 91.9% and stable planting survival rate was 98% , the tube seedlings survivaled retained all botany characteristics of Cynanchum wilfordii with exuberant growth and developed root system, harvest root fresh weight in- creasing 37% the following year.
