为研究B H K-21细胞的生长及代谢动力学特征,试验采用低密度静置培养方法,每天测定细胞生长密度和活力,并利用多参数生化分析仪测定培养液中Gluc、Lac、Gln和N H4+的含量,计算细胞指数生长期的比代谢速率。结果表明:BHK-21细胞生长曲线呈S型,最大增殖密度为50.3×104 upf·mL-1,倍增时间为21.1 h;对数生长期Gluc和Gln的比消耗速率为-3.79 mg·(106 cells)-1·d-1和-9.95μmol·(106 cells)-1·d-1,Lac和NH4+的比生成速率为2.81 mg·(106 cells)-1·d-1和6.30μmol·(106 cells)-1·d-1。

위연구B H K-21세포적생장급대사동역학특정,시험채용저밀도정치배양방법,매천측정세포생장밀도화활력,병이용다삼수생화분석의측정배양액중Gluc、Lac、Gln화N H4+적함량,계산세포지수생장기적비대사속솔。결과표명:BHK-21세포생장곡선정S형,최대증식밀도위50.3×104 upf·mL-1,배증시간위21.1 h;대수생장기Gluc화Gln적비소모속솔위-3.79 mg·(106 cells)-1·d-1화-9.95μmol·(106 cells)-1·d-1,Lac화NH4+적비생성속솔위2.81 mg·(106 cells)-1·d-1화6.30μmol·(106 cells)-1·d-1。
In order to research the grow th and metabolism kinetic characteristics of BHK-21 cells ,low cell densi-ty cultured method was used to determine cell density and vitality every day .The contents of Gluc ,Lac ,Gln , and NH4 + in medium were determined by multi-parameter biochemical analyzer ,and the specific metabolic rate were calculated .The results showed that the growth curve of BHK-21 cells was “S” type ,the largest density was 50 .3 × 104 upf·mL-1 and the doubling time was 21 .1 h .Specific consumption rate of Gluc and Gln were-3 .79 mg·(106 cells)-1 ·d-1 and -9 .95μmol·(106 cells)-1 ·d-1 in logarithmic phase ,and specific generation rate of Lac and NH4 + were 2 .81 mg·(106 cells)-1·d-1 and 6 .30 μmol·(106 cells)-1·d-1 .
