为获得高效转化木糖为乙醇的菌株,以木糖为唯一碳源,通过 TTC 平板显色法从自然环境中进行菌株的初筛,并分别对初筛菌株进行最佳发酵工艺条件的优化;再以相同发酵条件下乙醇产量为复筛指标筛选产醇率最高的菌株,进而对其发酵木糖产乙醇的工艺条件进行正交试验优化。结果表明:初筛共获得4株发酵木糖产乙醇菌株,其中 M3菌株的乙醇得率最高,为22.1%;试验对 M3个体形态以及菌落形态进行了初步鉴定,同时绘制了生长曲线;发酵木糖产乙醇的最佳发酵工艺为木糖浓度30 g/L,发酵时间48 h,初始 pH 5,发酵温度28℃,其乙醇得率达25.3%,比原条件下乙醇得率提高10.0%。

In order to obtain strains with high efficient of converting xylose into ethanol,TTC chromogenic screening plate was used to preliminarily screen ethanol-producing strains taking xylose as the single carbon source,and the fermentation process was optimized either.Then the process conditions of fermenting xylose to produce ethanol were optimized through an orthogonal test.The results showed that four strains were gained.Strain M3 got the highest ethanol yield,22.1%.The individual and colony forms of strain M3 were preliminarily identified and the growth curve was drawn;The optimized conditions of fermenting xylose to produce ethanol included xylose concentration 30 g/L,fermentation time 48 h,initial pH 5,fermentation temperature 28℃,ethanol yield 25.3%,10.0% higher than that under original conditions.
