建立了测定大鼠肝微粒体温孵体系中血根碱代谢产物二氢血根碱含量的高效液相色谱法。色谱柱为Shimadzu VP-ODSShim-pack(150mm×4.6mm,5μm);流动相为水:乙腈(37:63,V/V);流速1mL/min;柱温35℃;检测波长285nm;进样量10μL。结果显示50nmol/L和1000nmol/L二氢血根碱标准溶液的方法回收率分别为101.72%和98.82%;50、100、500nmol/,L标准溶液的日内RSD分别为2.26%、2.57%和3.02%,日间RSD分别为6.21%、4.33%和3.95%:二氢血根碱在25-1000nmol/L内与峰面积呈良好的线性关系(r=0.9996)。本分析方法快速简便,灵敏度高,重现性好,符合方法学验证的要求,可满足二氢血根碱在体外代谢中的定量研究。

An HPLC method for the quantitative determination of dihydrosanguinarine in rat liver microsomes incubation system was developed. Chromatography was performed on a Shimadzu VP - ODS column ( 150 mm ×4.6mm,5μm). Acetonitrile -water (63:37, V/V) as the mobile phase was used at 285 nm and 35 ℃. The recovery rate of 50 and 1000 nmol/L standard solution was 101.72% and 98.82% respectively. The relative standard deviation of 50,100 and 500 nmol/L standard solution for the within - day and between - day was 2.26% , 2.57%, 3.02% and 6.21%, 4.33% , 3.95% respectively. The calibration curve was linear within the range from 25 to 1000 nmol/L (r = 0. 9996 ). The results demonstrated that the method was simple, fast and suitable for the quantification of dihydrosanguinarine metabolised in vitro .
