通过检测发酵液无菌滤液对尖孢镰刀菌的抑制作用,研究了枯草芽孢杆菌SNUB16产生抗真菌物质的最佳条件及抗真菌物质的部分性质.结果表明,PDA培养液更有利于菌体的生长及抗菌物质的生成,在培养温度为30℃,初始pH值7.2,振荡速率为170r/min的条件下,培养60~72h时,无菌滤液对尖孢镰刀菌的抑菌作用最强.无菌滤液在50℃加热30min抗菌活性不受影响,而在60℃加热30min则抗菌活性下降;在100℃加热30min,抗菌活性丧失近一半,在121℃加热30min,抗菌活性完全丧失.在pH值6~7时抗菌物质的活性不受影响,但过酸和过碱会导致其抗菌活性逐步丧失,且过碱的影响大于过酸.蛋白酶K处理可使无菌滤液抗真菌活性部分丧失,推测枯草芽孢杆菌SNUB16产生的抗真菌物质除蛋白质类外,还包含其他非蛋白质类物质.

The cultural conditions and antifungal substances of Bacillus subtilis SNUB16 were investigated by determinating the inibition of cell-free fermentation filtrate to Fusarium ocrysporum. The results showed that PDA medium is favorable to cell growth and production of antifungal substance. After 60-72 hours culture at 30 ℃, initial pH of 7.2, oscillation rate of 170rpm, the cell-free fermentation filtrate has the highest antifungal activity to Fusarium oxysporum. The antifungal activity is not affected by heat at 50 ℃ for 30 min, but it decreases as the filtrate is heated at 60 ℃ for 30 min. Antifungal activity loses 50% when the filtrate is heated at 100 ℃ for 30 min, and antifudgal activity completely loses when the filtrate is heated at 121 ℃ for 30 min. The antifungal activity cannot be affected at the pH of 6 -7, but excess acid or alkaline can make its antifungal activity lose gradually,and the effect of excess alkaline is stronger than that of excess acid. The filtrate loses part of its antifungal activity when the filtrate is treated in presence of protease K,indicating that the primary antifungal substances produced by Bacillus subtilis SNUB16 are protein and non-protein.
