目的:探讨钩吻素子的基本理化性质及其水溶液稳定性。方法高效液相色谱法测定钩吻素子在不同介质中的平衡溶解度和表观油/水分配系数;考察钩吻素子在不同pH水溶液、不同温度下的稳定性。结果钩吻素子在纯水中的平衡溶解度为141 mg/mL ,在碱性介质中的平衡溶解度降低;酸性条件下钩吻素子的表观油/水分配系数对数值Lg P<-1,碱性条件下Lg P>1;4℃和25℃时钩吻素子水溶液在 pH 3~9稳定性良好,37℃时在强酸(pH 3)条件下稳定性较差。结论钩吻素子的水溶性较强,碱性条件下其亲脂性比酸性条件下高,高温条件下其强酸性水溶液稳定性较差。

목적:탐토구문소자적기본이화성질급기수용액은정성。방법고효액상색보법측정구문소자재불동개질중적평형용해도화표관유/수분배계수;고찰구문소자재불동pH수용액、불동온도하적은정성。결과구문소자재순수중적평형용해도위141 mg/mL ,재감성개질중적평형용해도강저;산성조건하구문소자적표관유/수분배계수대수치Lg P<-1,감성조건하Lg P>1;4℃화25℃시구문소자수용액재 pH 3~9은정성량호,37℃시재강산(pH 3)조건하은정성교차。결론구문소자적수용성교강,감성조건하기친지성비산성조건하고,고온조건하기강산성수용액은정성교차。
Objective To study the physicochemical proprieties of Koumine and the stability of Koumine solution . Methods High performance liquid chromatography (HPLC) are used to determine the equilibrium solubility and apparent oil/water partition coefficient of Koumine in different medium . The stability of Koumine solution was examined under the different pH and temperature . Results The equilibrium solubility of Koumine in water at 25 ℃ was 141 mg/mL and decreased in basic medium . The apparent oil/water partition coefficient Lg P was < -1 in neural and basic condition ,whereas Lg P was>1 in acidic condition . Between pH 3~9 ,the stability of Koumine solution was well sustained at 4 ℃and 25 ℃ . However ,the stability was significantly reduced under strong acidic condition (pH 3 .0) at 37 ℃ . Conclusion Koumine is highly soluble in water . It is more lipophilic in basic condition than in acidic condition . Under strong acidic and high temperature condition ,the stability of Koumine is poor .
