以云烟97、云烟87、Beinhart1000-1等11个烟草品种及N. glutinosa、N. plumbaginifolia为材料,采用SCoT分子标记技术,对上述11个烟草品种、3个烟草种间的遗传关系进行分析,并采用SCoT分子标记对10株N. plumbaginifolia和云烟87的杂交后代进行杂种鉴定。结果显示,对于云烟97,36条SCoT引物均扩增出了清晰的条带;分析11个烟草品种的PCR扩增条带,可筛选出的9条SCoT引物,共扩增出条带118条,其中多态性条带42条,多态性比例35.59%,各烟草品种间的相似系数均高于0.9;对2个野生种和普通烟草种进行扩增,筛选出的9条引物共扩增出163条带,其中多态性条带135条,多态性比例为82.82%,其中SP27号引物的多态性比例最高,达到了90.91%。10株杂交后代均可见父、母本特异条带,部分出现父母本不具有的特异条带。结果表明,SCoT引物不适于对普通烟草种内不同类型的遗传分析,可用于烟草种间的遗传关系的分析,也可用于远缘杂种的鉴定。

Genetic relationship of 11 tobacco varieties, e.g. Yunyan97, Yunyan87, Beinhart1000-1, and three interspecific tobacco varieties were analyzed using SCoT molecular marker technology. Ten hybrid offsprings of N. plumbaginifolia and Yunyan87 were identified. Results show that all 36 primers of the SCoT were amplified clear bands with Yunyan97 as template. Nine SCoT primers were selected from 11 tobacco varieties of PCR amplification, by which 118 bands were produced. Of them 42 polymorphic bands were found with polymorphism ratio of 35.59%. Similarity coefficients of all tobacco varieties were higher than 0.9. As for two wild species and an ordinary tobacco species, nine SCoT primers were selected from amplification results, by which 163 bands were produced, of them 135 polymorphic bands were found with polymorphism ratio of 82.82%. Primer SP27 showed the highest polymorphism of 90.91%. All 10 hybrid offsprings showed specific bands of male and female parent, while some of them showed new bands different from their parents. The above results indicated that existing SCoT primers were suitable for identifying genetic relationship between species and distant hybrids, but not suitable for varieties within species.
