目的:探讨甲状旁腺素对人甲状腺髓样癌细胞体外增殖抑制及凋亡作用。方法体外培养甲状腺髓样癌细胞株,经甲状旁腺素和甲状旁腺素受体单抗干预处理后,倒置相差显微镜下观察细胞生长状况,流式细胞仪检测细胞凋亡。结果倒置相差显微镜下细胞变化明显,各浓度的甲状旁腺素和甲状旁腺素受体单抗均分别能有效地抑制甲状腺髓样癌细胞增殖、诱导细胞凋亡,凋亡作用呈时间和浓度依赖。当甲状旁腺素浓度为2.0μmol/L、甲状旁腺素受体单抗浓度为1.0μmol/L时,对细胞凋亡作用显著(P<0.05),凋亡率分别为13.24%及20.78%。结论甲状旁腺素对人甲状腺髓样癌细胞增殖有抑制作用并能诱导其凋亡。

목적:탐토갑상방선소대인갑상선수양암세포체외증식억제급조망작용。방법체외배양갑상선수양암세포주,경갑상방선소화갑상방선소수체단항간예처리후,도치상차현미경하관찰세포생장상황,류식세포의검측세포조망。결과도치상차현미경하세포변화명현,각농도적갑상방선소화갑상방선소수체단항균분별능유효지억제갑상선수양암세포증식、유도세포조망,조망작용정시간화농도의뢰。당갑상방선소농도위2.0μmol/L、갑상방선소수체단항농도위1.0μmol/L시,대세포조망작용현저(P<0.05),조망솔분별위13.24%급20.78%。결론갑상방선소대인갑상선수양암세포증식유억제작용병능유도기조망。
Objective To explore the effects of parathyroid hormone on in vitro proliferation inhibition and apoptosis of cells of human medullary thyroid carcinoma. Methods Cell lines of medullary thyroid carcinoma were cultured in vitro and intervened by parathyroid hormone and monoclonal antibody of parathyroid hormone receptor.Cell growth was observed via inverted phase contrast microscope,and cell apoptosis was tested via flow cytometry. Results Cell changes were obvious under inverted phase contrast microscope,and parathyroid hormone and monoclonal antibody of parathyroid hormone receptor at all concentrations effectively inhibited cell proliferation and induced cell apoptosis of medullary thyroid carcinoma. The apoptosis was correlated with time and concentrations.When the concentration of parathyroid hormone was 2.0μmol/L,and the concentration of monoclonal antibody of parathyroid hormone receptor was 1.0μmol/L, the apoptosis effect was the most obvious(P<0.05),and the apoptosis rates were 13.24% and 20.78% respectively. Conclusion Parathyroid hormone has the effects of proliferation inhibition and apoptosis induction on cells of human medullary thyroid carcinoma.
