Hypericum perforatum extracts%Antidepressant%Behavior%Hippocampal neurons
目的:探究贯叶金丝桃提取物(hypericum perforatum extracts,HPE)对抑郁模型小鼠行为、海马神经元损伤的影响。方法用小鼠尾悬挂实验和强迫游泳实验对HPE的抗抑郁作用进行研究;将50只昆明种小鼠分为空白对照组,氟西汀组,HPE高(1.2g/kg)、中(0.60g/kg)、低剂量(0.20g/kg)组,采用长期不可预见性中等强度应激造成小鼠慢性应激抑郁模型,Nissl染色法观察海马CA1,CA3区神经元形态及锥体细胞数目。结果与空白对照组比较,HPE能显著缩短强迫游泳小鼠及尾悬吊小鼠的不动时间,并呈一定的剂量依赖性;与空白对照组比较,氟西汀组小鼠海马CA3区锥体细胞数量明显减少(P<0101),HPE高、中、低剂量组能明显增加海马CA3区锥体细胞数量。结论贯叶金丝桃提取物能改善小鼠的抑郁样行为,保护抑郁模型小鼠海马神经元损伤。

목적:탐구관협금사도제취물(hypericum perforatum extracts,HPE)대억욱모형소서행위、해마신경원손상적영향。방법용소서미현괘실험화강박유영실험대HPE적항억욱작용진행연구;장50지곤명충소서분위공백대조조,불서정조,HPE고(1.2g/kg)、중(0.60g/kg)、저제량(0.20g/kg)조,채용장기불가예견성중등강도응격조성소서만성응격억욱모형,Nissl염색법관찰해마CA1,CA3구신경원형태급추체세포수목。결과여공백대조조비교,HPE능현저축단강박유영소서급미현조소서적불동시간,병정일정적제량의뢰성;여공백대조조비교,불서정조소서해마CA3구추체세포수량명현감소(P<0101),HPE고、중、저제량조능명현증가해마CA3구추체세포수량。결론관협금사도제취물능개선소서적억욱양행위,보호억욱모형소서해마신경원손상。
Objective To observe the effects of hypericum perforatum extracts(HPE)on behavior,injury of hippocampal neurons in the depression mice model. Method Effects of HPE on behavior in depression model mice were observed by using the mouse tail suspension test and forced-swimming test models of depression;fifty Kunming mice were randomly divided into 5 groups,the control group,the model group and three HPE groups(the doses were 1.20g/kg、0.60g/kg、0.20g/kg respectively),The model was established by separation and chronic unpredictable mild stimulation,Cone cell and configuration of neuron in CA1,CA3 region of hippocampus were observed by Nissl. Result Comparing with the mice of the model group,immobility time of forced-swimming mice or tail suspension stress mice was significantly shortened by EHP in a dosedependent manner;comparing with the mice of control,numbers of cone cell in CA3 region of hippocampus were decreased obviously(P<0101)in the model group,HPE(1.20g/kg、0.60g/kg、0.20g/kg)could increase numbers of cone cell in CA3 region of hippocampus obviously(P<0.01,P<0.05,P<0.05). Conclusion HPE can improve the symptoms of the depression in the mouse model and protect injury of hippocampal neurons in the depression model mice.
